Abstract

2,3,7,8-Tetraklorodibenzo-p-dioksin (TCDD) adalah senyawa kimia paling toksik yang dihasilkan oleh kegiatan manusia dan memiliki salah satu efek sebagai karsinogenik. Dioksin dapat masuk ke dalam tubuh manusia melalui inhalasi, ingesti serta kontak kulit. Rosella ungu merupakan tanaman yang terkenal dalam bidang kesehatan. Yoghurt susu sapi fortifikasi ekstrak rosella ungu memiliki potensi sebagai antioksidan. Penelitian ini bertujuan untuk membuktikan potensi antioksidan yoghurt susu sapi fortifikasi ekstrak rosella ungu pada paparan dioksin yang diamati melalui kadar malondialdehid (MDA) dan histopatologi lambung. Penelitian ini dilakukan secara eksperimental menggunakan RAL. Hewan coba dibagi menjadi 5 kelompok yang terdiri dari kelompok kontrol negatif, kontrol positif (TCDD 200 ng/kg BB), kelompok perlakuan 1, 2, dan 3 (TCDD 200 ng/kg BB + yoghurt susu sapi fortifikasi rosella ungu konsentrasi 0,5%, konsentrasi 1%, dan konsentrasi 1,5%) diberikan per oral. Kadar MDA diukur dengan metode Thiobarbituric Acid (TBA) secara kuantitatif dengan uji statistik one way Analysis of Variance (ANOVA) dilanjutkan dengan uji Beda Nyata Jujur (BNJ) dengan taraf kepercayaan (p<0,05). Histopatologi lambung menggunakan metode pewarnaan Hematoksilin-Eosin (HE) yang diamati secara deskriptif. Hasil penelitian menunjukkan bahwa yoghurt susu sapi fortifikasi ekstrak rosella ungu 1% dapat mencegah kenaikan kadar MDA (p<0,05) dan memperbaiki kerusakan histopatologi pada lamina propria mukosa lambung tikus (Rattus norvegicus) yang dipapar TCDD. Kesimpulan penelitian ini adalah pemberian yoghurt susu sapi fortifikasi ekstrak rosella ungu dapat mencegah kenaikan kadar MDA dan memperbaiki kerusakan histopatologi lambung yang dipapar TCDD

English Abstract

2,3,7,8-Tetrachlorodiobenzo-p-dioxin (TCDD) is the most toxic chemistry compound produced by human activity and has one of the effects as carcinogenic. The route of TCDD to human by inhalation, ingestion and skin contact. Purple Roselle is a well-known plant in the medicine. Fortified yogurt with purple rosella extract has potential as an antioxidant . The purpose of this study was to prove the antioxidant potential of fortified yogurt with purple rosella extract on dioxin exposure observed by malondyaldehide (MDA) levels and histopathological of stomach. This study was occurred experimentally using RAL. Model animals were divided into 5 groups consisted of negative control group, positive control (TCDD 200 ng/kg BW), treatment groups 1, 2, and 3 (TCDD 200 ng/kg BW + fortified yogurt with purple rosella extract concentration 0,5%, concentration 1%, and concentration 1,5%) given orally. MDA level was measured by Thiobarbituric Acid (TBA) method as quantitatively using the one-way Analysis of Variance (ANOVA) statistical test followed by tukey’s honestly significant difference test (BNJ) with a level of trust (p<0.05). The histopathological of stomach was stained by Hematoxylin-eosin staining method which is observed descriptively. The result of this study showed that fortified yogurt with purple rosella extract 1% could prevent the increase level of MDA (p<0,05) and repair histopathological damage to the lamina propria mucosa of stomach rat (Rattus norvegicus) exposed to TCDD. The conclusion of this study was the administration of yogurt with purple rosella extract could prevent the increase level of MDA and repair histopathological damage to the stomach exposed to TCDD.
