Abstract

Dioksin merupakan senyawa toksik dan berdampak buruk pada sistem kardiovaskular. Yoghurt susu sapi fortifikasi ekstrak rosella ungu berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian yoghurt susu sapi fortifikasi ekstrak rosella ungu terhadap paparan dioksin terhadap kadar Malondialdehida (MDA) dan histopatologi jantung. Penelitian ini dilakukan selama 12 hari dan bersifat eksperimental dengan menggunakan Rancangan Acak Lengkap. Tikus (Rattus norvegicus) sebanyak 30 ekor dibagi menjadi 5 kelompok, yaitu kontrol negatif, kontrol positif menggunakan TCDD, perlakuan 1 dengan dosis 200 ng/kgBB TCDD dan 1 mL yoghurt susu sapi fortifikasi ekstrak rosella ungu 0,5%, perlakuan 2 dengan dosis 200 ng/kgBB TCDD dan 1 mL yoghurt susu sapi fortifikasi ekstrak rosella ungu 1,0%, dan perlakuan 3 dengan dosis 200 ng/kgBB TCDD dan 1 mL yoghurt susu sapi fortifikasi ekstrak rosella ungu 1,5%. Kadar MDA jantung diukur dengan menggunakan metode Thiobarbituric Acid (TBA) dan histopatologi jantung dengan menggunakan metode pewarnaan Hematoksilin Eosin (HE). Kadar MDA jantung kemudian dilakukan analisis dengan menggunakan one way Analysis of Variance (ANOVA). Histopatologi jantung diwarnai dengan menggunakan pewarnaan Hematoksilin Eosin dilakukan analisis secara deskriptif. Hasil penelitian menunjukkan bahwa yoghurt susu sapi fortifikasi ekstrak ungu dapat mencegah kenaikan kadar MDA signifikan (p<0.05) dan mencegah kerusakan histopatologi jantung akibat paparan TCDD. Konsentrasi 1,5% fortifikasi ekstrak rosella ungu pada yoghurt susu sapi merupkan konsentrasi yang terbaik. Kesimpulan penelitian ini dalah pemberian fortifikasi ekstrak rosella ungu pada yoghurt susu sapi dapat mencegah kenaikan kadar MDA dan menghambat terjadinya kerusakan histopatologi jantung akibat paparan TCDD.

English Abstract

Dioxin is compounds considered to be toxic and TCDD exposure also adversely affects the cardiovascular system. Purple rosella fortified cow's milk yogurt has potential as an antioxidant. This study aims to determine the effect of purple rosella fortified yogurt on dioxin exposure observed from the levels of Malondialdehyde (MDA) and histopathological of the heart. This research is experimental by using a completely randomized design (CRD). 30 rats (Rattus norvegicus) were divided into 5 groups, namely negative control, positive control (using TCDD), treatment 1 with a dose of 200 ng/kg TCDD and 1 mL of yogurt fortified purple rosella 0.5%, treatment 2 with a dose of 200 ng/kgBB TCDD and 1 mL of yogurt fortified purple rosella 1.0%, and treatment 3 with a dose of 200 ng / kgBB TCDD and 1 mL of yogurt fortification of rosella purple 1.5%. Heart MDA levels were measured using the Thiobarbituric Acid (TBA) and histopathology heart using the Haematoxylin Eosin (HE) staining method. MDA levels of the heart were then analyzed using one-way Analysis of Variance (ANOVA). Histopathology heart using hematoxylin-eosin staining was analyzed descriptively. MDA levels of the heart were then analyzed using one way Analysis of Variance (ANOVA). Histopathology of the heart was stained using Hematoxylin Eosin staining using descriptive analysis. The results showed that the fortified purple extract of cow's milk yogurt could prevent a significant increase in MDA levels (p <0.05) and prevent heart myocyte cells that were experiencing necrosis due to TCDD exposure. The concentration of 1.5% fortification of purple rosella extract in cow's milk yogurt is the best concentration. The conclusion of this study is the fortification of purple rosella extract fortification in cow's milk yogurt can prevent the increase in MDA levels and inhibit the occurrence of necrosis due to TCDD exposure.
