Abstract
Diabetes mellitus adalah penyakit kronis yang disebabkan oleh berkurangnya produksi hormon insulin. Selain itu, dapat disebabkan oleh peningkatan kadar gula darah. Yoghurt susu kambing dan bekatul beras merah memiliki kandungan antioksidan (peptida bioaktif, tokoferol, oryzanol, dan flavonoid) dan serat tinggi, sehingga baik untuk terapi diabetes mellitus. Penelitian ini dilakukan untuk melihat pengaruh pemberian yoghurt susu kambing fortifikasi tepung bekatul beras merah terhadap kadar MDA dan histopatologi pankreas hewan model diabetes mellitus induksi streptozotosin. Penelitian ini menggunakan Rancangan Acak Lengkap dengan 20 ekor tikus putih yang terbagi menjadi lima kelompok, yaitu kontrol negatif tanpa perlakuan apapun. Sementara itu, kontrol positif, perlakuan 1, 2, dan 3 diinduksi streptozotosin secara intraperitoneal dosis 45 mg/kgBB. Setelah diinkubasi selama 3 hari, perlakuan 1, 2, dan 3 diberi terapi yoghurt susu kambing fortifikasi tepung bekatul beras merah dengan volume pemberian 1 mL/kgBB, 2 mL/kgBB, dan 3 mL/kgBB selama 14 hari. Kadar MDA diukur menggunakan spektrofotometer dan histopatologi pankreas diwarnai dengan pewarnaan HE. Analisa data kadar MDA dianalisa dengan uji ANOVA dan uji Tukey α = 0,05, sedangkan histopatologi pankreas secara deskriptif. Hasil penelitian menunjukkan terapi yoghurt susu kambing fortifikasi tepung bekatul beras merah volume 1 mL/KgBB, 2 mL/KgBB, dan 3 mL/KgBB dapat menurunkan kadar MDA dan memperbaiki histopatologi pankreas yang ditandai dengan regenerasi pada sel beta pulau Langerhan’s pankreas. Kesimpulan pada penelitian ini yoghurt susu kambing fortifikasi tepung bekatul beras merah dengan 2 mL/KgBB adalah volume pemberian terbaik dan menghasilkan hasil yang efektif untuk terapi diabetes mellitus dengan menurunkan kadar MDA dan memperbaiki histopatologi pankreas yang ditandai dengan regenerasi pada sel beta pulau Langerhan’s pankreas
English Abstract
Diabetes mellitus is a chronic disease caused by the deficiency of insulin hormone. In addition, it can cause the increasing amount of blood sugar levels in despression condition. Goat milk yoghurt and red rice bran flour have high antioxidant (bioactive peptide, tocopherol, oryzanol, and flavonoid) and fiber, therefore it might be good for diabetes mellitus treatment. The propose of this research was to observe the effect of goat milk yoghurt fortification of red rice bran flour towards MDA levels and pancreatic histopathology of diabetes mellitus rats (Rattus norvegicus) models induced by streptozotosin. This research used a completely randomized design (CRD) with 20 rats which weight around 130-200 grams and divided into five groups, including negative control without any treatment. Meanwhile positive control, treatment 1, 2, and 3 were intraperitoneal injection of 45 mg/KgBW streptozotosin. After 3 days incubation, treatment 1, 2, and 3 received 1 mL/KgBW, 2 mL/KgBW, and 3 mL/KgBW goat milk youghurt with fortification of red rice bran flour respectively for 14 days. MDA levels were measured by spectrophotometry and pancreatic histophatology was stained by Hematoxylin eosin. Data of MDA levels were analyzed using ANOVA test and Tukey test α = 0,05, while pancreatic histophatology were analyzed descriptively. The result showed that 1 mL/KgBW, 2 mL/KgBW, and 3 mL/KgBW of goat milk yoghurt with fortification of red rice bran decreased pancreatic MDA levels and repair the pancreatic histophatology indicated by regeneration in beta cell of Langerhan’s island. The conclusion of this research is goat milk yoghurt with fortification of red rice bran with volume of 2 mL/KgBW was effective for theraphy diabetes mellitus by decreasing MDA levels and repairing the pancreatic histophatology indicated by regeneration in beta cell of Langerhan’s island.
