Abstract

Dioksin adalah kelompok senyawa yang bersifat racun (toksik) dan merupakan faktor pemicu kanker. Dioksin paling beracun adalah 2,3,7,8- tetrachloro-dibenzo-p-dioksin (TCDD). Dioksin dapat mengakibatkan gangguan kesehatan karena sifat persisten dan dapat terbioakumulasi dalam lemak tubuh. Kasein yoghurt susu kambing berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian yoghurt susu kambing terhadap paparan dioksin yang diamati dari kadar malondialdehida (MDA) dan histopatologi jantung. Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL). Tikus putih (Rattus norvegicus) dibagi menjadi 6 kelompok, yaitu kontrol negatif, kontrol kasein (kasein yogurt susu kambing dosis 600 mg/kgBB), kontrol positif (TCDD dosis 100 ng/kgBB), perlakuan 1 (kasein yogurt susu kambing 300 mg/kgBB dan TCDD 100 ng/kg BB), perlakuan 2 (kasein yogurt susu kambing 600 mg/kgBB dan TCDD 100 ng/kgBB), dan perlakuan 3 (kasein yogurt susu kambing 900 mg/kg BB dan TCDD 100 ng/kgBB). Kadar MDA jantung diukur menggunakan metode Thiobarbituric Acid (TBA) dengan spektrofotometri UV-Vis pada panjang gelombang 530 nm dan histopatologi jantung menggunakan metode pewarnaan Hematoksilin Eosin (HE). Kadar MDA jantung dianalisis dengan one way Analysis of Variance (ANOVA) tingkat kepercayaan α=5% dan analisis histopatologi jantung secara deskriptif. Hasil penelitian menunjukkan kasein yoghurt susu kambing dapat mencegah kenaikan kadar MDA signifikan (p<0,01) dan mencegah sel miosit jantung yang mengalami nekrosis akibat paparan TCDD. Dosis kasein yoghurt susu kambing 900mg/kgBB merupakan dosis terbaik. Kesimpulan penelitian yaitu pemberian kasein yoghurt susu kambing dapat mencegah kenaikan kadar MDA dan mencegah sel miosit jantung yang mengalami nekrosis akibat paparan TCDD.

English Abstract

Dioxin are a group of compounds that are poisonous (toxic) and it was a factor to trigger cancer. The most toxic dioxin was 2,3,7,8-tetrachloro-dibenzop- dioxin (TCDD). Dioxin can cause health problems due to the nature of persistent and can bioaccumulative in the fat body. Casein goat milk yoghurt potential as an antioxidant. Research aimed was to determine effect of casein goat milk yoghurt to dioxin exposure were observed in the levels of malondialdehida (MDA) and histopathology picture of heart. It was experimental study by used a Completely Randomized Desing (CRD). Rats (Rattus norvegicus) were devide into 6 groups, the each namely were: negative control, casein control (Casein goat milk yoghurt 600mg/kgBW), positive control (TCDD 100ng/kgBW), treatment 1 (Casein goat milk yoghurt 300mg/kgBW and TCDD 100ng/kgBW), treatment 2 (Casein goat milk yoghurt 600mg/kg BW and TCDD 100ng/kgBW), and treatment 3 (Casein goat milk yoghurt 900mg/kgBW and TCDD 100ng/kgBW). Heart MDA levels measured by used Thiobarbituric Acid (TBA) with UV-Vis spectrophotometry at 530nm wavelength and histopathology picture of heart by used staining method hematoxylin and Eosin (HE). MDA levels was analyzed by one way Analysis of Variance (ANOVA) confidence level α=5% and histopatology of heart analyzed by descriptive. Results showed casein goat milk yoghurt can prevent increase of MDA levels highly significantly (p<0,01) and prevent myosit cell from necrosis caused by TCDD exposure. Casein goat milk yoghurt 900mg/kgBW is the best dose. Conclusion of research was casein goat milk yoghurt can prevent increase of MDA levels and myosit cell from necrosis caused by TCDD exposure.
