Abstract

Dioksin merupakan senyawa toksik yang termasuk dalam senyawa organoklorin bersifat sangat reaktif dan menghasilkan radikal bebas yang dapat merusak sel lambung. 2,3,7,8-Tetrachlorinedibenzo-p-dioksin (TCDD) merupakan jenis dioksin yang memiliki toksisitas paling tinggi. Kasein yoghurt susu kambing memiliki potensi sebagai antioksidan. Penelitian ini bertujuan mengetahui efek preventif pemberian yoghurt susu kambing terhadap paparan dioksin yang diamati kadar malondialdehida (MDA) dan gambaran histopatologi lambung. Penelitian bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL). Tikus putih (Rattus norvegicus) dibagi 6 kelompok, yaitu kelompok normal, kontrol kasein (kasein yoghurt susu kambing dosis 600 mg/kg BB), kontrol positif (TCDD dosis 100 ng/kg BB), perlakuan 1 (kasein yoghurt susu kambing 300 mg/kg BB dan TCDD 100 ng/kg BB), perlakuan 2 (kasein yoghurt susu kambing 600 mg/kg BB dan TCDD 100 ng/kg BB), dan perlakuan 3 (kasein yoghurt susu kambing 900 mg/kg BB dan TCDD 100 ng/kg BB). Kadar MDA lambung diukur menggunakan metode uji Thiobarbituric Acid (TBA) dengan pengukuran spektrofotometri UV-Vis panjang gelombang 530 nm dan gambaran histopatologi lambung menggunakan pewarnaan Hematoksilin Eosin (HE) diamati berupa kerusakan sel mukosa lambung. Kadar MDA lambung dianalisis menggunakan uji statistik one way ANOVA, α=5% dan analisis histopatologi lambung diolah secara deskriptif. Hasil penelitian menunjukkan kasein yoghurt susu kambing dengan dosis 600 dan 900 mg/kgBB merupakan dosis terbaik dalam mencegah kenaikan kadar MDA lambung dari tikus putih (Rattus norvegicus) terpapar TCDD secara signifikan (p<0,01), dan dosis 900mg/kgBB mencegah terjadi erosi sel mukosa lambung. Kesimpulan penelitian yaitu pemberian kasein yoghurt susu kambing mampu mencegah kenaikan kadar MDA dan terjadi erosi sel mukosa lambung akibat TCDD

English Abstract

Dioxins are toxic compounds belonging to organochlorine compounds are highly reactive and produce free radicals that can damage stomach cells. 2,3,7,8- Tetrachlorinedibenzo-p-dioxin (TCDD) is the type of dioxin that has the highest toxicity. Casein goat milk yogurt has the potential as an antioxidant. This study aims to determine the preventive effect of goat milk yoghurt on exposure to dioxin observed levels of malondialdehyde (MDA) and gastric histopathology. Experimental research using Completely Randomized Design (RAL). The white rats (Rattus norvegicus) were divided into 6 groups, the normal group, casein control (goat milk yoghurt dose 600 mg / kg BW), positive control (TCDD dose 100 ng / kg BW), treatment 1 (goat milk yoghurt 300 mg / kg BW and TCDD 100 ng / kg BW), treatment 2 (goat milk yoghurt 600 mg / kg BW and TCDD 100 ng / kg BW), and treatment 3 (goat milk yoghurt 900 mg / kg BW and TCDD 100 ng / kg BW). Gastric MDA levels were measured using the Thiobarbituric Acid (TBA) assay method with 530 nm wavelength UV-Vis spectrophotometric measurement and gastric histopathology using Hematoxylin Eosin (HE) staining observed in the form of gastric mucosal cell damage. Gastric MDA levels were analyzed using one way ANOVA statistical test, α = 5% and gastric histopathology analysis was descriptively processed. The results showed that goat milk yoghurt with dose of 600 and 900 mg / kgBW was the best dose in preventing the increase of MDA level of gastric from white rat (Rattus norvegicus) exposed TCDD significantly (p <0,01), and dose 900mg / kgBB prevented erosion of gastric mucosal cells. The conclusion of this research is giving goat milk goat yoghurt able to prevent the increase of MDA level and erosion of gastric mucosal cells caused by TCDD
