Abstract

Dioksin merupakan senyawa kimia paling toksik yang dihasilkan oleh kegiatan manusia dan memiliki salah satu efek sebagai reprotoksik. Jenis dioksin yang memiliki toksik paling tinggi adalah 2,3,7,8-Tetraklorodibenzo-p-dioksin (TCDD). Kasein yoghurt susu kambing memiliki potensi sebagai antioksidan. Penelitian ini bertujuan untuk membuktikan potensi antioksidan yoghurt susu kambing pada paparan dioksin yang diamati melalui kadar malondyaldehide (MDA) dan gambaran histopatologi testis. Penelitian ini dilakukan secara eksperimental menggunakan RAL. Hewan coba dibagi menjadi 6 kelompok yang terdiri dari kelompok normal, kelompok kontrol kasein (kasein yogurt susu kambing 600 mg/kg BB), kelompok positif (TCDD 100 ng/kg BB), kelompok perlakuan 1 (kasein yoghurt susu kambing 300 mg/kg BB dan TCDD 100 ng/kg BB), kelompok perlakuan 2 (kasein yoghurt susu kambing 600 mg/kg BB dan TCDD 100 ng/kg BB) dan kelompok perlakuan 3 (kasein yoghurt susu kambing 900 mg/kg BB dan TCDD 100 ng/kg BB). Kadar MDA diukur dengan metode Thiobarbituric Acid (TBA) menggunakan spektrofotometri UV-Vis panjang gelombang 530 nm secara kuantitatif dengan uji statistik one way Analysis of Variance (ANOVA) dilanjutkan dengan uji Beda Nyata Jujur (BNJ). Gambaran histopatologi testis menggunakan metode pewarnaan Hematoksilin-Eosin (HE) diamati secara deskriptif sel sperma dalam tubulus seminiferus. Hasil penelitian menunjukkan bahwa kasein yoghurt susu kambing dapat mencegah kenaikan kadar MDA dan pengurangan kepadatan sel spermatozoa akibat paparan TCDD secara signifikan (p<0,05). Dosis 900 mg/kg BB adalah dosis terbaik dalam mencegah kenaikan kadar MDA dan pengurangan kepadatan sel spermatozoa. Kesimpulan penelitian ini adalah pemberian kasein yoghurt susu kambing dapat mencegah kenaikan kadar MDA dan pengurangan kepadatan spermatozoa akibat paparan TCDD

English Abstract

Dioxin is the most toxic chemistry compound produced by human activity and has one of the effects as the reprotoxic. The type of dioxin that has the highest toxicity is 2,3,7,8-Tetraklorodibenzo-p-dioksin (TCDD). Goat milk yoghurt casein has a potential as antioxidant. The purpose of this study was to prove the antioxidant potential of goat milk yoghurt on dioxin exposure observed by malondyaldehide (MDA) levels and histopathological image of testicle. This study was occurred experimentally using RAL. Model animals were divided into 6 groups consisted of normal group, casein control group (goat milk yoghurt casein 600 mg/kg BW), positive group (TCDD 100 ng/kg BW), treatment group 1 (goat milk yoghurt casein 300 mg/kg BW and TCDD 100 ng/kg BW), treatment group 2 (goat milk yoghurt casein 600 mg/kg BW and TCDD 100 ng/kg BW) and treatment group 3 (goat milk yoghurt casein 900 mg/kg BW and TCDD 100 ng/kg BW). MDA level was measured by Thiobarbituric Acid (TBA) method using spectrophotometry UV-Vis with wavelength of 530 nm quantitatively with one way Analysis of Variance (ANOVA) statistic test method, continued by Honest Significant Difference test. The histopathological image of testicle was stained by Hematoxylin-eosin staining method, and sperm cells in seminiferous tubules were observed descriptively. The result of this study showed that goat milk yoghurt casein could prevent the increase level of MDA and decrease the density of spermatozoa cells as the effect of TCDD exposure significantly (p<0,05). The dose of 900 mg/kg BW was the most best dose to prevent MDA level to increase and the density of spermatozoa cells to decrease. The conclusion of this study was the administration of goat milk yoghurt casein could prevent the increase level of MDA and decrease the density of spermatozoa cells as the effect of TCDD exposure.
