English Abstract
Uric acid disease (gout) is a disease caused by the accumulation of uric crystals in the joint that will cause inflammation in the long term, as well as tissue damage resulting in pain. Uric acid disease is caused by hyperuricemia condition i.e. excessive uric acid in the blood (serum uric acid (sUA)> 6.8 mg/dL). Medical treatment has a high risk of side effects such as nausea, skin rashes, diarrhea, and potentially renal failure. The use of natural medicine is considered safer because it has the minor possibility of side effects. Moringa leaf, pandan leaf and red ginger are known to have bioactive compounds performing as antioxidants. Formulation of the three ingredients might perform as anti-hyperuricemia and anti-inflammation. The purpose of this study is to determine antioxidant performance of supplements made of formulation of the three ingredients extracted using the solvent of water and 70% ethanol. Phase I of the study was the production of supplements through the process of encapsulation with 2% chitosan coating agent and drying using freeze drying method, with a series of activities: extraction, formulation, optimization and encapsulation. The extraction process of moringa leaf and pandan leafwas performedin two methods, namely maceration using 70% ethanol solvent and infusion using water solvent. Red ginger rhizome was extracted using a pressure of 2000kg/cm2, then the resulting dregs were each macerated using 70% ethanol solvent and water solvent. The three viscous extractswere then formulated using a linear program assisted by POM QM version 3 with the purpose (Z) of minimized IC50 value. The ratio solution resulted from the extract formulation of moringa leaf, pandanleaf and red ginger was 70%:20%:10%. Formulationresult of the viscous extract was then optimized using One Factor Response Surface Methodology (RSM). The optimization result states that the ratio of2%chitosan coating agent and extract volume was 1:3. Phase II of this study was the test of water extract supplements (WES) and ethanol extract supplements(EES) to the hyperuricemic male rats (induced by potassium oxonate) and inflammatory male mice (induced byuric monosodium crystal) using Randomized Complete Design (RCD). The result of Phase IIshows that WES and EES were able to perform as anti-hyperuricemia and anti-inflammation so that this supplement can be an alternative treatment of hyperuricemia and inflammation.
