Abstract

Fibrosis hepar menyebabkan 1,5 juta kematian setiap tahun. Kematian ini sebagian besar disebabkan oleh stadium akhir fibrosis berupa sirosis dan kanker hepar. Transplantasi hepar merupakan satu-satunya terapi pada stadium akhir ini. Namun terapi ini memerlukan biaya yang mahal dan sulit mencari organ donor. Di sisi lain, kulit kacang tanah (Arachis hypogea L.) selama ini dianggap sebagai limbah, padahal banyak mengandung flavonoid. Jenis flavonoid terbanyak yang dikandung adalah luteolin. Luteolin mampu menghambat aktivasi Hepatic Stellate Cell (HSC) yang memproduksi kolagen dan meningkatkan MT I/II yang berperan dalam proses regenerasi hepar. Tujuan penelitian ini adalah membuktikan pengaruh ekstrak kulit kacang tanah terhadap penurunan deposisi kolagen hepar mencit model fibrosis hepar. Penelitian ini merupakan true experimental design yang menggunakan metode Randomized Post-test Only Controlled Group Design. Kami menggunakan mencit Balb/c jantan yang dibagi menjadi 5 kelompok; 1) kelompok kontrol negatif, tidak diberi CCl4 maupun ekstrak kulit kacang tanah, 2) kelompok kontrol positif, diberi CCl4 tanpa ekstrak kulit kacang tanah, 3) kelompok P1, diberi CCl4 dan ekstrak kulit kacang tanah 15 mg/KgBB, 4) kelompok P2, diberi CCl4 dan ekstrak kulit kacang tanah 30 mg/KgBB, dan 5) kelompok P3, diberi CCl4 dan ekstrak kulit kacang tanah 60 mg/KgBB. Hasil penelitian menunjukkan adanya penurunan jumlah kolagen hepar secara mikroskopik pada mencit yang diinduksi fibrosisi hepar dan diberi ekstrak kulit kacang tanah. Sehingga dapat disimpulkan bahwa ekstrak kulit kacang tanah mampu menurunkan deposisi kolagen pada hepar mencit Balb/c model fibrosis hepar.

English Abstract

Liver fibrosis causes 1.5 million deaths per year. It is largely causes by the last stage of liver fibrosis such as cirrhosis and liver cancer. Liver transplantation is the only one therapy at this stage. But this therapy needs expensive cost. It also very hard to find donor accordance. In other side, peanut hull (Arachis hypogea L.) is considered as rubbish and unutilized, while it contains flavonoid so much. The kind of flavonoid which mostly contained in peanut hull is luteolin. Luteolin can inhibit activation of Hepatic Stellate Cell (HSC) that produce collagen. It also increase MT I/II that increase in regenerating hepatic cell. The purpose of this experiment is to prove the effect of peanut hull extract at decreasing collagen deposition in liver fibrosis model mice. This reseacrh is true experimental design and use Randomized Post-test Only Controlled Group Design. We use male Balb/c mice and divided into 5 group; 1) negative control, does not inducted, 2) positive control, inducted by CCl4 without peanut hull extract, 3) P1, inducted by CCl4 and 15 mg/Kg BW peanut hull extract, 4) P2, inducted by CCl4 and 30 mg/Kg BW peanut hull extract, 5) P3, inducted by CCl4 and 60 mg/Kg BW peanut hull extract. The result shows the decrease of collagen in liver microscopicly in mice inducted liver fibrosis and peanut hull extract. We conclude that peanut hull extract decrease collagen deposition in Balb/c mice liver fibrosis.
