Abstract

Limbah jerami padi memiliki jumlah yang cukup besar yaitu sekitar 50,19 juta ton tahun 2016. Jerami padi mengandung lignoselulosa yang terdiri dari 32,1% selulosa, 24% hemiselulosa, dan 18% lignin. Lignin sendiri mengandung sejumlah komponen fenolik terutama ferulic, p-coumaric, syringic, vanillic, dan p-hydroxybenzoic yang dapat diturunkan menjadi senyawa aromatik seperti BTX (benzene, toluene, dan xylene), fenol, dan vanilin. Proses degradasi lignin menjadi senyawa-senyawa tersebut dilakukan menggunakan mikroorganisme salah satunya jamur pelapuk coklat yaitu Serpula lacrymans. Hasil degradasi yang diamati berupa total gula reduksi (TGR), total soluble phenol (TSP), pH, dan susut berat kering. Proses selanjutnya berupa optimasi proses ekstraksi menggunakan metode response surface method (RSM) terdiri dari dua faktor yaitu volume pelarut etanol dan lama ekstraksi. Respon berupa kadar vanilin dan kandungan vanilin. Hasil solusi optimal akan diidentifikasi senyawa vanilin menggunakan LC-ESI-MS/MS. Hasil uji parameter proses degradasi menunjukkan nilai rata-rata tertinggi adalah lama inkubasi 21 hari. Hasil optimal proses ini berupa volume pelarut sebesar 40,28 ml dan lama ekstraksi 249,11 menit untuk kedua respon. Kadar vanilin yang diperoleh sekitar 6,20% dan yield vanilin sekitar 2,59 μg/g dengan desirability 0,839 atau 83,9%. Identifikasi senyawa vanilin dengan LC-ESI-MS/MS didapatkan waktu retensi 2,53 menit namun tidak dapat diukur karena jumlahnya terlalu kecil

English Abstract

The potential of Rice straw wastes were 50.19 million tons in 2016. Rice straw is one of the lignocelluloses feedstock contains 32.1% cellulose, 24% hemicellulose, and 18% lignin. Lignin contains a number of phenolic components especially ferulic, p-coumaric, syringic, vanillic, and p-hydroxybenzoic which can be degraded into aromatic compounds such as BTX (benzene, toluene and xylene), phenols and vanillins. The degradation process of lignin into these compounds is carried out by microorganisms one of them is brown rot fungi, Serpula lacrymans. The observed parameters of degradation were total sugar reduction (TGR), total soluble phenol (TSP), pH, and weight loss. Ethanol volume and extraction time were optimized by response surface method (RSM) based on the highest TSP in order to obtain vanillin content and yield vanillin. The results indicated that for 21 days incubating rice straw resulted the highest TSP and TGR, the lowest pH while the highest weight loss values were obtain from 28 days incubation. The optimum result of this process was ethanol volume of 40.28 ml and extraction time of 249.11 min which produced vanillin with the content of 6.20% and the yield vanillin of 2.59 μg / g. The desirability value was 0.839 or 83.9%, which used to simultaneously optimize the multiple responses and represents the closeness of both responses to its ideal value. Vanillin compound was identified by LC-ESI-MS/MS showed 2.53 minute retention time but there was no detectable amount of vanillin.
