Abstract

Perkembangan teknologi yang semakin pesat tidak dapat dipisahkan dari peningkatan paparan radikal bebas yang menimbulkan berbagai penyakit seperti kanker. Hal ini menjadi tantangan untuk terus mencari dan mengembangkan sumber-sumber antioksidan baru. Senyawa fenol terbukti memiliki aktvitas antioksidan yang potensial dan dapat ditemukan dengan mudah dalam berbagai produk berbasis bahan alam, salah satunya pada buah juwet (Syzygium cumini). Buah juwet yang semakin sulit diperoleh menjadi hambatan tersendiri, sehingga perlu dipertimbangan pemanfaatan dari bagian tumbuhan juwet yang lain seperti biji dan daun. Alternatif ini perlu disertai dengan dilakukannya analisis penetapan kadar total fenol dari masing-masing bagian tumbuhan tersebut agar diperoleh acuan perbandingan yang bermanfaat bagi efikasi dan efisiensi pemanfaatan tumbuhan juwet kedepannya. Tujuan dari penelitian ini adalah untuk mengetahui seberapa besar kadar total fenol dari ekstrak etanol 96% buah, biji, dan daun tumbuhan juwet (Syzygium cumini) dan apakah terdapat perbedaan yang signifikan diantara ketiganya. English Abstract

The rapid development of technology can not be separated from the increase in exposure to free radicals that cause various diseases such as cancer. This is a challenge to look for and develop a new source of antioxidants. Phenol compounds are believed to have a potential antioxidant activity and can be found easily in a variety of natural-based products, one of which is on juwet fruit (Syzygium cumini). Juwet fruit which is increasingly difficult to obtain becomes its own obstacle, so it needs to be considered the utilization of other parts of the juwet plant such as seed and leaf. This alternative needs to be accompanied by an analysis to determining the total phenolic content of each part of the plant in order to obtain a comparative reference that is useful for the efficacy and efficiency of juwet plants. This study aims to determine the amount of total phenolic content of 96% ethanolic extract in the fruit, seed, and leaf of juwet (Syzygium cumini) and, whether there were significant differences between each part. The determination of total phenolic content was carried out by the Folin-Ciocalteu method and UV-Vis spectrophotometer instrument. Validation method performed several parameters included specificity test (λ = 747.00 nm), linearity (y = 0.1032 x - 0.0732; r = 0.99), LOD (0,839 ppm), LOQ (2,796 ppm), accuracy (98,095 - 109,333%), and precision (2,112% - 4,385%). The total phenol content of 96% ethanol extract of fruit, seed, and leaf of juwet (Syzygium cumini) obtained was 24,09 ± 0.576 mg GAE/g, 503,01 ± 9.215 mg GAE/g, and 329,60 ± 20,378 mg GAE/g, where the One Way Anova test and Post-Hoc test yielded p < 0.05 which showed a significant difference between the three groups.
