Abstract

Perubahan pola makan dan gaya hidup yang tidak sehat di masyarakat menyebabkan meningkatnya prevalensi hipertensi.Hipertensi terjadi akibat peningkatan curah jantung dan atau resistensi perifer. Resistensi perifer utamanya diperankan sel endotel dan sel otot polos pembuluh darah. Endotel berfungsi untuk menghasilkan NO sebagai vasodilator otot polos. Zat toksik seperti ROS, kerusakan sel dapat memicu proses inflamasi yang menghasilkan TNF-⍺. TNF-⍺ juga dapat memicu keluarnya ROS yang menyebabkan disfungsi endotel, akibatnya terjadi penurunan produksi NO dan otot polos pembuluh darah akan cenderung mengalami vasokonstriksi dan berujung pada hipertensi. Ekstrak daun ciplukan (Physalis angulata L.) mengandung fisalin yang bermanfaat sebagai anti-inflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun ciplukan terhadap kadar TNF-⍺ serum tikus wistar betina model hipertensi. Jumlah sampel yang digunakan sebanyak 25 ekor, yang dibagi menjadi 5 kelompok. Kelompok kontrol negatif yang tidak diberi perlakuan, kontrol positif yaitu model hewan hipertensi (diinduksi L-NAME 75 mg/kgBB secara subkutan), Perlakuan 1 yaitu model hewan hipertensi disertai pemberian ekstrak daun ciplukan 500 mg/kgBB, Perlakuan 2 dengan dosis ekstrak 1500 mg/kgBB, dan Perlakuan 3 dengan dosis ekstrak 2500 mg/kgBB. Pengukuran kadar TNF-⍺ serum dilakukan dengan metode ELISA. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak daun ciplukan dapat menyebabkan penurunan kadar, namun penurunan yang dihasilkan tersebut tidak signifikan secara statistik. Semakin tinggi dosis ekstrak ciplukan, semakin rendah kadar TNF-⍺ serum, hubungan antara dosis ekstrak dengan kadar TNF-⍺ serum pada korelasi sedang. Perlu dilakukan evaluasi dosis dan rute pemberian agar menghasilkan hasil yang signifikan.

English Abstract

Changes in eating patterns and unhealthy lifestyles in society have led to an increase in the prevalence of hypertension. Hypertension occurs due to increased cardiac output and/or peripheral resistance. Peripheral resistance is mainly played by endothelial cells and vascular smooth muscle cells. Endothelial functions to produce NO as a smooth muscle vasodilator. Toxic substances such as ROS, cell damage can trigger an inflammatory process that produces TNF-⍺. TNF-⍺ can also trigger the release of ROS which causes endothelial dysfunction, as a result there is a decrease in NO production and the smooth muscle of blood vessels will tend to experience vasoconstriction and lead to hypertension. Ciplukan leaf extract (Physalis angulata L.) contains fisalin which is useful as an anti-inflammatory. This study aims to determine the effect of ciplukan leaf extract on serum TNF-⍺ levels of female hypertensive models of Wistar rats. The number of samples used was 25 individuals, which were divided into 5 groups. The negative control group that was not treated, the positive control was a hypertensive animal model (L-NAME 75 mg/kgBW induced subcutan), Treatment 1 was an animal model of hypertension accompanied by 500 mg/kgBB of ciplukan leaf extract, treatment 2 with an extract dose of 1500 mg/kgBB, and treatment 3 with an extract dose of 2500 mg/kgBB. Measurement of serum TNF-⍺ levels was carried out by the ELISA method. The results of this study indicate that administration of ciplukan leaf extract can cause a decrease in levels, but the resulting decrease is not statistically significant. The higher the ciplukan extract dose, the lower the TNF-⍺ serum level, the relationship between extract dose and serum TNF-⍺ level was in moderate correlation. It is necessary to evaluate the dose and route of administration in order to produce significant results.
