Abstract
osis berpotensi tercemar atau bercampur dengan bahan dari spesies daging lainnya seperti babi yang tidak diharapkan oleh konsumen. Deteksi berbasis DNA metode PCR primer spesies spesifik babi ND5 (NADH dehydrogenase subunit 5) memiliki beberapa kelebihan, yaitu sederhana, spesifik dan sensitif. Tahapan preparasi pada deteksi berbasis DNA, yaitu metode isolasi. Metode isolasi DNA pada olahan daging telah banyak digunakan namun membutukan waktu yang lama, biaya tinggi, dan keahlian khusus. Penelitian ini bertujuan membandingkan tiga metode isolasi DNA, yaitu metode alkali, urea dan garam pada sosis dengan modifikasi tanpa menggunakan enzim proteinase K dan nuklease. Metode ini memiliki beberapa kelebihan di antaranya ekonomis, cepat, mudah dan menghasilkan konsentrasi dan kemurnian DNA yang cukup baik. Primer spesies spesifik berhasil mengamplifikasi sampel sosis babi dan daging babi dengan ukuran 227 bp. Metode alkali menjadi rekomendasi karena cepat, mudah dilakukan, ekonomis, menghasilkan konsentrasi dan kemurnian DNA yang cukup baik serta pita DNA yang tebal.
English Abstract
Sausages have the potential to be contaminated or mixed with ingredients from other meat species such as pork that are not expected by consumers. DNA-based detection of primary PCR method for specific species of ND5 pigs (NADH dehydrogenase subunit 5) has several advantages, which is simple, specific and sensitive. First stages of preparation for DNA-based detection are isolation method. DNA isolation methods in processed meat have been widely used but consume a lot of time, high costs, and special expertise. This study aims to compare three DNA isolation methods, they were alkali, urea and salt methods in sausages with modification without using the enzyme proteinase K and nucleases. This method has several advantages including being economical, fast, easy and produce sufficient concentration and purity of DNA. Specific species primers successfully amplified samples of pork sausages and pork meat wit a size of 227 bp. The alkaline method is a recommendation because it is fast, easy to do, economical, produces sufficient DNA concentration and purity and very thick DNA bands.
