Abstract
Musa balbisiana Colla merupakan pisang liar yang mempunyai banyak biji di dalam buahnya. Daun pisang batu mengandung senyawa epigallocatechin gallate (EGCG) yang bertindak sebagai antioksidan dan obat terapi kanker. Teknik ekstraksi kavitasi tekanan negatif digunakan untuk ekstraksi EGCG karena teknik ini tidak menggunakan panas. Faktor penting yang mempengaruhi efisiensi teknik kavitasi adalah tekanan dan rasio pelarut: zat terlarut. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh perbedaan tekanan dan rasio pelarut: zat terlarut terhadap kadar dan rendemen EGCG ekstrak daun pisang batu. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) pola faktorial dengan dua faktor. Faktor 1 adalah tekanan (-0,03 MPa; -0,05 MPa; dan - 0,07 MPa) dan faktor 2 adalah rasio pelarut : zat terlarut (20:1 ml/g; 22:1 ml/g; dan 24:1 ml/g). Hasil ekstraksi dilakukan analisis kandungan total flavonoid, kadar dan rendemen EGCG, serta aktivitas antioksidan. Berdasarkan hasil penelitian, rasio pelarut: zat terlarut berpengaruh nyata terhadap kandungan total flavonoid, sedangkan tekanan dan interaksi antar kedua faktor tidak berpengaruh nyata terhadap kandungan total flavonoid. Larutan ekstrak dengan kandungan total flavonoid tertinggi diperoleh pada kondisi tekanan -0,05 MPa dengan rasio pelarut : zat terlarut 24 : 1 ml/g. Larutan ekstrak ini memiliki kandungan total flavonoid sebesar 210 µgQE/ml ekstrak dengan kadar EGCG sebesar 16,4 mg/l. Rendemen EGCG yang diperoleh sebesar 0,029%, serta nilai IC50 sebesar 96,47 ppm yang termasuk ke dalam aktivitas antioksidan kuat
English Abstract
Musa balbisiana Colla is a wild banana that has many seeds in its fruit. Batu banana leaves contain the compound epigallocatechin gallate (EGCG) which is capable of acting as an antioxidant and cancer therapy. The negative pressure cavitation extraction technique is used for EGCG extraction because it does not use heat. Important factors that influence the results of efficiency cavitation techniques are pressure and ratio solvent: solute. The purpose of this study was to determine the effect of pressure differences and ratio solvent: solute on the concentration and yield of EGCG Batu banana leaf extract. This study uses a Randomized Block Design (RBD) factorial pattern with two factors. Factor 1 is pressure (-0.03 MPa; -0.05 MPa; and -0.07 MPa) and factor 2 is the ratio of solvent: solute (20: 1 ml / g; 22: 1 ml / g; and 24: 1) ml / g). The extraction results analyze for total flavonoid content, concentration, and yield of EGCG, and antioxidant activity. Based on the results of this study, the ratio solvent: solute has a significant effect on the total content of flavonoids, while the pressure and interactions between the two factors have no significant effect on the total content of flavonoids. The extract solution with the highest total flavonoid content obtains under pressure conditions of -0,05 MPa with a ratio solvent: solute 24: 1 ml/g. This extract solution has a total flavonoid content of 210 µgQE/ml extract with EGCG concentration of 16,4 mg/l. The yield of EGCG obtained was 0,029%, and the IC50 value was 96,47 ppm which include in the strong antioxidant activity
