Abstract
Tanaman jeruk lemon (Citrus limon) merupakan salah satu tanaman obat yang memiliki banyak manfaat karena mengandung sumber vitamin C, kalium and kalsium. Permasalahan utama dari pertumbuhan tanaman lemon yaitu adanya penyakit diplodia yang disebabkan oleh cendawan Botryodiplodia theobromae. Pengendalian biologis seperti penggunaan bakteri endofit digunakan sebagai agen untuk mengendalikan patogen dan meningkatkan pertumbuhan tanaman lemon. Penelitian ini dilakukan untuk mendapatkan bakteri endofit sebagai agen biokontrol yang mampu menghasilkan enzim kitinase dan mendegradasi dinding sel kitin pada jamur patogen. Tahapan pada penelitian ini adalah isolasi bakteri endofit dari tanaman jeruk lemon di BALITJESTRO, karakterisasi bakteri secara makroskopis dan mikroskopis, seleksi bakteri penghasil kitinase, uji antagonis terhadap jamur Botryodiplodia theobromae, uji aktivitas enzim kitinase, dan karakterisasi enzim kitinase yang dihasilkan. Berdasarkan hasil penelitian diperoleh 12 isolat yang berhasil diisolasi, 7 diantaranya memiliki kemampuan menghambat jamur patogen dan 3 diantaranya yaitu K3a, D dan D6 mampu menghasilkan enzim kitinase serta menghambat pertumbuhan jamur Botryodiplodia theobroame. Hasil uji antagonis secara mikroskopis isolat K3a, menunjukan bahwa hifa dari jamur patogen mengalami abnormalitas. Sedangkan secara makroskopis, isolat bakteri endofit memiliki nilai indeks kitinolitik tertinggi sebesar 1,5 cm dan daya antagonis mencapai nilai 93%. Aktivitas enzim kitinase dari isolat K3a dengan konsentrasi koloidal kitin 0,5% yang optimal terjadi pada fase log atau jam ke- 44, yaitu sebesar 0,220 Unit/ml. Suhu optimal dari enzim kitinase diperoleh pada 50oC dengan nilai aktivitas 0,205 U/ml dan pH 7 dengan nilai aktivitas 0,212 Unit/ml.
English Abstract
Lemon (Citrus limon) is one of the plants that has potential as a traditional medicine because lemon is source of Vitamin C, calium and calsium. The primary problem associated with growing lemon plants is the occurance of diplodia disease, which is caused by Botryodiplodia theobromae. Biology methode such as used endophytic bacteria can be used to control pathogen and promote the growth of the lemon plants. This research aims to determine the presence of endophytic bacteria producing chitinase enzyme as biocontrol in lemon plant and inhibit growth of fungal Botryodiplodia theobromae. This research was conducted with several steps are isolation of endophytic bacteria from lemon plant in BALITJESTRO, identification of endophytic bacteria include macroscopic and microscopic examinition, selection the bacteria that produce chitinase enzyme, antagonist test with fungal pathogen, chitinase assay of chitinase enzyme, and characterization of chitinase enzyme (pH and temperature). Based on the result, 12 isolates endophyte were successfully isolated, 7 isolates have ability to inhibit the patoghen fungi. 3 isolates (K3a, D and D6) showed ability to produced chitinase enzyme and inhibit growth of Botryodiplodia theobromae. The microscopic result that hyphae of fungal pathogen was abnormality, whilst based on macroscopic result it was indicated that K3a produced clear zone with highest chitinolytic index was 1.5 cm and inhibition against fungal pathogen was ranged 93%. The crude enzyme activity of K3a with 0.5% colloidal chitin was 0.220 U/ml in log phase with the optimum temperature was 50oC with 0.205 U/ml and the optimum pH 7.0 with a high number of enzyme activity of 0.212 U.ml.
