Abstract

Radikal Bebas Yang Dihasilkan Oleh Asap Rokok Telah Diketahui Dapat Mengganggu Fungsi Organ Reproduksi Termasuk Jaringan Testis. Efek Lokal Maupun Sistemik Dari Paparan Asap Rokok Dapat Dijelaskan Melalui Mekanisme Stres Oksidatif Dan Inflamasi. Biji Anggur (Vitis Vinifera) Diketahui Mengandung Proantosianidin, Flavonoid, Katekin, Dan Epikatekin Yang Memiliki Aktivitas Sebagai Antioksidan Penelitian Bertujuan Untuk Mengetahui Pengaruh Pemberian Ekstrak Biji Anggur (Vitis Vinifera) Terhadap Ekspresi Interleukin-1-Beta (IL-1β) Dan Gambaran Histopatologi Testis Tikus Yang Diberi Paparan Asap Rokok. Penelitian Menggunakan Rancangan Acak Lengkap Dengan 20 Ekor Tikus Putih (Rattus Norvegicus) Strain Wistar Jantan Umur 12 Minggu Dan BB 175-200 Gram Dibagi Menjadi 5 Kelompok: K1 (Kelompok Negatif), K2 (Kelompok Positif Yang Dipaparan Asap Rokok Selama 14 Hari Pertama), P1 (Kelompok Perlakuan Pasca Pemaparan Asap Rokok Yang Diberikan Ekstrak Air Biji Anggur (Vitis Vinifera) Dosis 0,9 Mg/200 Gram BB Tikus Selama 14 Hari), P2 (Dosis 2,7/200 Gram BB Tikus), Dan P3 (Dosis 5,4/200 Gram BB Tikus). Hasil Penelitian Menunjukkan Bahwa Pemberian Ekstrak Biji Anggur (Vitis Vinifera) Dapat Menurunkan Ekspresi IL-1β Testis Tikus Pada Kelompok Terapi Dibandingkan Dengan Kelompok Kontrol Positif. Penurunan Ekspresi IL-1β Yang Paling Rendah Ditemukan Pada Kelompok P3 (Dosis 5,4 Mg/200 Gram BB Tikus) Sebesar 48% Dibandingkan Dengan Kelompok Kontrol Positif Yaitu Sebesar 0.979±0.009. Pemberian Ekstrak Biji Anggur (Vitis Vinifera) Dengan Dosis 5,4 Mg/200 Gram BB Tikus Juga Paling Efektif Dalam Memperbaiki Kerusakan Sel Sertoli, Sel Leydig Dan Diameter Tubulus Seminiferus Yang Ditandai Dengan Perkembangan Sel Spermatogenik Pada Jaringan Testis Tikus

English Abstract

Free Radicals Produced By Cigarette Smoke Can Disturb The Function Of Reproductive Organs, Including Testis. Local And Systemic Influences Of Cigarette Smoke Exposure May Be Through The Occurrence Of Oxidative Stress And Inflammation. Grape (Vitis Vinifera) Seed Contains Of Porantocyanidin, Flavonoid, Catechin, And Epycathecin Which Have Antioxidant Activity. Research Is Aimed To Understand The Influence Of Treatment By Grape (Vitis Vinifera) Seed Extract On The Expression Of Interleukin-1-Beta (IL-1β) And Testis Histopathology Appearence Of Rat Exposed By Cigarette Smoke. Research Design Is Complete Random Planning. The Object Of Research Is 20 Wistar Strain Rat (Rattus Norvegicus) With 12 Weeks Old And 175-200 Grams Body Weight. Rat Are Assigned Into 5 Groups Which Are K1 (Negative Group), K2 (Positive Group Exposed By Cigarette Smoke For First 14 Days), P1 (Treatment Group Which Is After Cigarette Smoke Exposure, Given With The Liquid Extract Of Grape (Vitis Vinifera) Seed For 14 Days At Dose 0.9 Mg/200 Grams Body Weight), P2 (Similar To P1 But At Dose 2.7 Mg/200 Grams Body Weight), And P3 (Similar To P1 But At Dose 5.4/200 Grams Body Weight). Result Of Research Indicates That The Treatment Of Grape (Vitis Vinifera) Seed Extract Can Reduce The Expression Of IL-1β Of Rat Testis In Therapy Group If Compared To Positive Control Group. The Lowest Reduction Of IL-1β Expression Is Found At P3 (At Dose 5.4 Mg/200 Grams Body Weight) By 48 % If Compared To Positive Control Group Which Reaches 0.979±0.009. Treating Mouse With Grape (Vitis Vinifera) See Extract At Dose 5.4 Mg/200 Grams Body Weight Is The Most Effective Method To Repair The Damages On Sertoli Cells, Leydig Cells, And Tubulus Seminiferus Diameter Which Is Signed By The Development Of Spermatogenic Cells In Rat Testis
