Abstract
Penelitian secara in vivo perlu memperhatikan komposisi zat gizi yang terkandung dalam diet yang diberikan sebagai suatu standarisasi diet pada hewan coba. Standarisasi diet tersebut digunakan untuk menurunkan variasi atau bias pada interpretasi hasil penelitian. Diet standar yang umum diberikan pada hewan coba tikus meliputi 2 jenis diet yaitu diet normal standar PAR-S dan diet normal standar AIN-93M. Keduanya memiliki kandungan zat gizi yang berbeda, sehingga bila penerapan standar diet tidak dikendalikan dapat menyebabkan bias pada penelitian. Penelitian ini bertujuan untuk mengukur dan menganalisis perbedaan kadar LDL (Low Density Lipoprotein) serum pada tikus yang diberi diet normal standar PAR-S dan diet normal standar AIN-93M. Jenis penelitian ini adalah studi eksperimental menggunakan randomized control group posttest design. Subjek penelitian ialah tikus Wistar jantan yang dipelihara di Laboratorium Biosains Universitas Brawijaya selama 12 minggu. Sampel dipilih dengan cara RAL (Rancangan Acak Lengkap) dibagi dalam dua kelompok dengan jumlah total 32 tikus. Variabel yang diukur dalam penelitian ini ialah kadar LDL. Hasil penelitian menunjukkan bahwa kadar LDL pada kedua kelompok tidak berbeda secara signifikan (Uji T, p=0.531; p>0.05). Rata-rata kadar LDL pada kelompok diet normal standar PAR-S 25.1 ± 15.4 mg/dL dan pada kelompok diet normal standar AIN-93M 24.7 ± 6.61 mg/dL. Kesimpulan pada penelitian ini adalah tidak terdapat perbedaan yang signifikan pada kadar LDL karena dipengaruhi oleh metabolisme lipoprotein, pengaruh hormonal, dan efek diet yang diberikan.
English Abstract
The in vivo studies should be observed about nutrient content of diet that could be used to standardize studies among laboratories. The aim of standardization of test diets for laboratory animals was to reduce the variation interpretation of results among experiments. In general, there are two type of standard diet for laboratory animals are the standard normal diet PAR-S and standard normal diet AIN-93M. Both have different nutrient composition. If the diet is not controlled, it can provide variation in the study. This study was conducted to measure and analyze differences of serum LDL in rats that receive the standard normal diet PAR-S and standard normal diet AIN-93M. This research was true experimental using randomized control group posttest design. Subjects were male Rattus norvegicus strain wistar that placed in Biosains Laboratory of Brawijaya University for 12 weeks. Sample was carried out by completely random sampling with 32 rats for all group. Serum LDL levels were measured by indirect method. The average of serum LDL levels in rats that receive standard normal diet PAR-S are 25.1 ± 15.4 mg/dL and in rats that receive standard normal diet AIN-93M are 24.7 ± 6.61 mg/dL. The conclusion is there was not significant difference of serum LDL at two groups (T test, p=0.531; p>0.05). It’s because side effect of metabolism lipoprotein in rats, hormone, and diet effects.
