Abstract
Karies gigi merupakan salah satu penyakit gigi dan mulut yang umum terjadi pada berbagai lapisan masyarakat. Upaya kuratif untuk menangani masalah ini tidak bisa menurunkan insiden, bahkan meningkat seiring dengan usia. Jadi upaya preventif yang terbaik adalah pengobatan alternatif untuk menurunkan insiden karies. Buah pinang (Areca catechu Linn) adalah salah satu tanaman obat tradisional digunakan. Buah pinang (Areca catechu Linn) mengandung alkaloid , flavonoid dan tannin yang memiliki efek antibakteri. Penelitian ini bertujuan untuk mengetahui efek ekstrak etanol buah pinang (Areca catechu Linn) sebagai antibakteri terhadap Streptococcus mutans secara in vitro. Penelitian ini dilakukan dengan metode agar well diffusion. Sampel diperoleh dari isolat bakteri di Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Brawijaya. Konsentrasi ekstrak yang digunakan adalah 3,125% ; 6,25% ; 12,5% ; 25% ; 50%, kontrol positif chlorhexidine gluconate 0,2% dan kontrol negatif aquades. Kemudian diinkubasi selama 2x24 jam, 370C. Perhitungan diameter zona hambat dengan menggunakan jangka sorong dengan ketelitian 0.05 mm. Analisis data menunjukkan perbedaan bermakna antara berbagai konsentrasi ekstrak etanol buah pinang (Areca catechu Linn) terhadap pertumbuhan Streptococcus mutans (p<0,05). Uji regresi linier sederhana menunjukkan adanya pengaruh konsentrasi ekstrak etanol buah pinang (Areca catechu Linn) dalam menghambat pertumbuhan Streptococcus mutans. Uji korelasi menunjukkan adanya hubungan signifikan antara pemberian ekstrak etanol pinang (Areca catechu Linn) terhadap pertumbuhan Streptococcus mutans. Kesimpulan pada penelitian ini yaitu ekstrak etanol buah pinang (Areca catechu Linn) mempunyai efek antibakteri terhadap Streptococcus mutans dengan diameter zona hambat terbesar 14.1625 milimeter pada konsentrasi ekstrak etanol buah pinang (Areca catechu Linn) 50% dan diameter zona hambat terkecil 11.95 milimeter pada konsentrasi ekstrak etanol buah pinang (Areca catechu Linn) 3.125%.
English Abstract
Dental caries is one of the most common tooth and mouth disease occurs in various society. Curative effort to handle this problem cannot decrease the number of the incidents, more over it is increased along with age. So, the best preventive effort is alternative medication to decrease the number of caries incidents. Areca fruit (Areca catechu Linn) is one of the many traditional medicinal plants used. Areca fruit (Areca catechu Linn ) contains alkaloid, flavonoid and tannin which has an antibacterial effect. This study aims to determine the effect of areca fruit ethanol extract (Areca catechu Linn) as an antibacterial against Streptococcus mutans in vitro. This study was done using agar well diffusion method. The samples were obtained from bacterial isolates provided by the Microbiology Laboratory of Medical Faculty of Brawijaya University. The extract concentration used were 3,125% ; 6,25% ; 12,5% ; 25% ; 50%, positive control that use chlorhexidine gluconate 0,2% and negative control that use aquadest. Which then incubated for 2X24 hours, 370C. Measuring the diameters of inhibiting zone using calipers with the precision of 0.05 mm. Data analysis showed the significant differences among various concentration of the extract to the growth of Streptococcus mutans ( p < 0.05). Simple linear regression test showed the presence of the influence of the concentration of areca fruit ethanol extract (Areca catechu Linn) in inhibiting the growth of Streptococcus mutans. The correlation test showed a significant relation between the granting of areca fruit ethanol extract (Areca catechu Linn) against the growth of Streptococcus mutans. The conclusion is that areca fruit ethanol extract (Areca catechu Linn) has an antibacterial effect against Streptococcus mutans with the biggest inhibitory zone diameter 14.1625 millimeter on areca fruit ethanol extract (Areca Catechu Linn) concentration 50% and the smallest inhibitory zone diameter 11.95 millimeter on areca fruit ethanol extract (Areca Catechu Linn) concentration 3.125%.
