Abstract
Efek hiperglikemia, peningkatan asam lemak bebas dan resistensi insulin pada diabetes melitus mengakibatkan disfungsi endotel dan pembentukan ROS. Tingginya ROS menyebabkan rendahnya produksi NO. Ketidakseimbangan antara ROS dengan antioksidan endogen dapat diantisipasi dengan adanya antioksidan eksogen. Albumin merupakan salah satu antioksidan eksogen yang banyak terkandung dalam ekstrak ikan gabus. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak ikan gabus (Channa striata) terhadap kadar Nitric Oxide (NO) pada tikus Rattus norvegicus jantan strain Wistar model diabetes melitus. Penelitian ini merupakan studi eksperimental yang terdiri dari 5 kelompok yaitu kontrol negatif (P0) yaitu kelompok normal, kontrol positif (P1) yaitu kelompok DM, kelompok perlakuan 1 (P2) yaitu kelompok DM + ekstrak ikan gabus 3 ml/kg BB, kelompok perlakuan 2 (P3) yaitu kelompok DM + ekstrak ikan gabus 6 ml/kg BB dan kelompok perlakuan 3 (P4) yaitu kelompok DM + ekstrak ikan gabus 9 ml/kg BB. Hasil uji statistik Oneway ANOVA menunjukkan signifikansi (Sig=0,92>0,05), artinya tidak ada pengaruh pemberian ekstrak ikan gabus terhadap kadar NO. Dengan demikian disimpulkan bahwa pemberian ekstrak ikan gabus tidak berpengaruh secara signifikan untuk meningkatkan kadar NO pada tikus Rattus norvegicus jantan strain Wistar model diabetes melitus.
English Abstract
Effects of hyperglycemia, increased free fatty acids and insulin resistance in diabetes mellitus resulting in endothelial dysfunction and the formation of ROS. The high ROS leads to low production of NO. Imbalance between ROS and endogenous antioxidants may be anticipated in the presence of exogenous antioxidants. Albumin is one of the many exogenous antioxidants contained in snakehead fish extract. This study aimed to determine the effect of snakehead fish extract (Channa striata) to the levels of Nitric Oxide (NO) in Diabetes Mellitus male rats (Rattus norvegicus strain Wistar). This research was an experimental study that consists of 5 groups: negative control (P0) was normal group, the positive control (P1) was group of DM, treatment group 1 (P2) was group of DM + snakehead fish extract 3 ml/kg BW, treatment group 2 (P3) was group of DM + snakehead fish extract 6 ml/kg BW and treatment group 3 (P4) was group of DM + snakehead fish extract 9 ml/kg BW. Oneway ANOVA statistical test results showed significant (Sig = 0.92> 0.05), meaning that snakehead fish extract had no effect on levels of NO (p=0,92). It is concluded that snakehead fish extract doesn’t significantly increase the levels of NO in Diabetes Mellitus male rats (Rattus norvegicus strain Wistar).
