Abstract
Glutathione merupakan salah satu antioksidan yang berperan dalam menjaga kualitas semen dan kemampuan fertilisasi spermatozoa. Tujuan penelitian ini adalah menganalisis dan mengevaluasi pengaruh glutathione pada pengencer dasar Ringer’s terhadap kualitas semen ayam buras selama simpan dingin dan menguji fertilitas telur yang diinseminasi menggunakan semen dalam bahan pengencer Ringer’s dengan penambahan glutathione pada simpan dingin. Penelitian dilaksanakan di Instalasi Ternak Unggas dan Laboratorium Reproduksi Sekolah Tinggi Penyuluhan Pertanian (STPP) Malang. Penelitian tahap I, menggunakan Rancangan Acak kelompok (RAK) pola tersarang dengan 4 perlakuan pengencer yaitu 1) Ringer’s tanpa glutathione, 2) Ringer’s + 0,1 mM glutathione, 3) Ringer’s+0,5 mM glutathione dan 4) Ringer’s+1 mM glutathione. Parameter kualitas semen selama simpan dingin meliputi motilitas individu, viabilitas, dan abnormalitas spermatozoa yang diamati pada jam ke-0, 4, 8, 24, 32, dan 48 pasca simpan dingin. Analisis data menggunakan Analisis of Variance (ANOVA) dan Duncan Multiple Range Test (DMRT). Penelitian tahap II, uji fertilitas telur menggunakan Rancangan Acak Lengkap (RAL) 3 perlakuan pengencer semen dan analisis data menggunakan chi-square. Hasil penelitian menunjukkan bahwa perlakuan penambahan glutathione dalam pengencer Ringer’s menunjukkan perbedaan yang sangat nyata (P<0,01) terhadap kualitas semen. Hasil terbaik ditunjukkan pada perlakuan Ringer’s +0,5 mM glutathione pada lama simpan jam ke-8, dengan rataan motilitas 62±2,91%, viabilitas 79,26±2,68% dan abnormalitas 13,66±0,31%, dan hasil uji fertilitas telur 42%. Kesimpulan pada penelitian ini adalah penambahan antioksidan glutathione dalam pengencer Ringer’s mampu mempertahankan kualitas spermatozoa ayam buras selama simpan dingin pada konsentrasi terbaik 0,5 mM Glutathioe selama 8 jam simpan dingin. Inseminasi Buatan menggunakan semen segar dengan pengencer Ringer’s menunjukkan persentase fertilitas telur terbaik. Inseminasi Buatan dengan menggunakan semen dingin belum menunjukkan persentase fertilitas telur yang baik meskipun telah ditambahkan glutathione 0,5 mM sebagai antioksidan.
English Abstract
Glutathione is one of the antioxidants that play a role in maintaining of semen quality and ability of sperm fertilization.The purpose of this study was to analyze and evaluate the influence of glutathione in the basic diluent Ringer's during cold storage and test to egg fertility are inseminated using semen in diluents Ringer's solution with the addition of glutathione stored in cold temperatures. The research was conducted at Poultry Installation and Reproductive Laboratory of Sekolah Tinggi Penyuluhan Pertanian (STPP) Malang. The first phase, using Random Design Group (RAK) pattern of nested 4 treatment diluent: 1) Ringer's without glutathione, 2) Ringer's + 0,1 mM glutathione, 3) Ringer's + 0,5 mM glutathione and 4) Ringer's + 1 mM Glutathione. Semen quality parameters during cold storage included individual motility, viability, and spermatozoa abnormalities observed at 0, 4, 8, 24, 32 and 48 post cold storage hours. Analysis of data using Analysis of Variance (ANOVA) and continued with Duncan Multiple Range Test (DMRT). The second phase, test the fertility of eggs with a completely randomized design (CRD) 3 treatments diluent and data analysis used chi-square non parametric analysis. The results of the study showed that the addition of glutathione in Ringer's diluent showed very significant differences (P <0,01) on semen quality. The best results are shown in Ringer's treatment on a long +0,5 mM glutathione save hours to 8, with a mean of 62 ± 2,91% motility, viability of 79,26 ± 2,68% and 13,66 ± 0,31% abnormalities and egg fertility 42% The conclusion in this study was the addition glutathione in Ringer’s diluent can survive semen quality, with best concentration was 0,5 mM glutathione in Ringer's diluent and cold storage until 8 hours. Artificial insemination using fresh semen with Ringer's diluent shows the best percentage of fertility of eggs. Artificial insemination using cold semen has not demonstrated a good percentage of fertility of eggs despite the addition of 0,5 mM glutathione as an antioxidant.
