Abstract
Mutasi yang terjadi pada gen hepatocyte nuclear factor 1 (Hnf1) menjadi salah satu faktor dalam mekanisme patogenesis penderita diabetes melitus tipe 2 (DMT2). Penggunaan nutrisi alami diperlukan untuk memperbaiki kerusakan tingkat gen pada penderita DMT2. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian protein CSN1S2 dari susu kambing Ethawah dalam menghambat mutasi gen Hnf1 exon 2 dan 4 menggunakan tikus model DMT2. Penelitian ini dilakukan dengan menggunakan tikus (Rattus norvegicus strain Wistar) yang dibagi menjadi 2 kelompok utama dengan 3 ulangan yaitu kontrol dan kelompok diabetes yang telah diinduksi streptozotocin (STZ) pada dosis 25 mg/kg BB. Perlakuan protein CSN1S2 diberikan pada 3 dosis yaitu 375 mg/kg BB; 750 mg/kg BB; 1500 mg/kg BB. Produk isolasi DNA total dari organ pankreas diamplifikasi untuk mendapatkan sekuen target yaitu gen Hnf1 exon 2 dan 4. Hasil amplifikasi disekuensing lalu dilakukan analisis menggunakan BioEdit. Hasil penelitian menunjukkan bahwa penggunaan STZ dapat menginduksi mutasi yang mengarah pada DMT2 pada gen Hnf1 exon 4 yaitu insersi T99 à C99. Mutasi tersebut dapat dihambat oleh protein CSN1S2 pada penggunaan dosis 375 dan 750 mg/kg, sedangkan pada dosis 1500 mg/kg BB terjadi mutasi baru yaitu insersi T90 à G90 dan insersi A415 à T415. Sehingga penggunaan dosis aman untuk kelompok DMT2 adalah 375 dan 750 mg/kg BB.
English Abstract
Mutations in the hepatocyte nuclear factor 1 gene (Hnf1) become one of the factors which cause type 2 diabetes mellitus (T2DM), by using CSN1S2 protein from PE goat milk is expected to improve the defect at genomic level in T2DM patients. The aim of this study to analyze the effect of protein CSN1S2 Ethawah goat milk for minimize the mutations in Hnf1 exon 2 and 4 of rat (Rattus norvegicus Wistar strain) T2DM model. This research focus in two main groups model with three repetitions, there are normal group and diabetic group which induced by streptozotocin (STZ). All groups were given CSN1S2 protein in three doses: 375 mg/kg BW, 750 mg/kg BW and 1500 mg/kg BW. After all treatment, Hnf1 gene was extracted from pancreas then amplified with specific primer. The results of amplification are sequenced using ABL PRIMS 3700 DNA Sequencer and analyzed using BioEdit. The results showed the induction of STZ can cause mutations that lead to T2DM in Hnf1 gene exon 4 and does not occur in exon 2. The type of mutation is insertion T99 à C99. Mutations in exon 4 were minimized after given CSN1S2 protein at 375 and 750 mg/kg BW doses. Whereas at doses 1500 mg/kg BW cause new mutation, that is insertion T90 à G90 and insertion A415 à T415. Accordingly, the safe dosage for T2DM is 375 and 750 mg/kg BW
