Abstract
Obesitas merupakan salah satu masalah kesehatan yang ada di Indonesia. Daun kelor (Moringa oleifera) sebagai salah satu tanaman yang banyak tumbuh di daerah tropis memiliki manfaat salah satunya sebagai antiobesitas. Namun pengaruh daun kelor belum diketahui pada zebrafish. Zebrafish merupakan model yang baik untuk diet induced obesity (DIO) dan dalam meneliti profil lipid sebab proses pencernaan makanan dan lipogenesis pada zebrafish mirip dengan pada manusia. Penelitian ini bertujuan untuk menilai efek kelor terhadap kadar kolesterol pada zebrafish dengan DIO. Penelitian eksperimental dilakukan dengan post test only control group design. Zebrafish yang digunakan adalah zebrafish dewasa (5 mpf) dan kelor yang digunakan adalah ekstrak daun kelor yang dilarutkan dalam ethanol. Perlakuan terhadap zebrafish terdiri dari kontrol negatif (non DIO), kontrol positif (DIO), dosis A (DIO + ekstrak daun kelor 0,56 ppm), B (DIO + ekstrak daun kelor 1,12 ppm) dan C (DIO + ekstrak daun kelor 2,24 ppm) dengan jumlah sampel tiap perlakuan delapan ekor. Variabel yang diukur adalah kadar kolesterol total, kadar HDL, dan kadar LDL/VLDL dari lemak visceral zebrafish. Hasil dari penelitian ini menunjukkan adanya perbedaan secara signifikan dari kontrol non DIO ke kontrol DIO, dan pemberian ekstrak daun kelor pada zebrafish dengan DIO menunjukkan efek perbaikan profil lipid berupa penurunan kolesterol total (p=0.002), peningkatan konsentrasi HDL (p=0.00), serta penurunan kadar LDL/VLDL (p=0.00)pada dosis 0,56 ppm. Pada dosis 1,12 ppm dan 2,24 ppm tidak terdapat perbaikan profil lipid yang signifikan.
English Abstract
Obesity is one of many problems rising in Indonesia. One can become obese as the lipid cells proliferate and undergo hypertrophy. Kelor (Moringa oleifera) is known to grow amply in tropical areas with as ample benefits, one of which is its anti obesity effect. But this effect is yet to be studied in zebrafish. Zebrafish is a promising diet induced obesity (DIO) model and a good model to use for lipid research for its similarity of metabolism organ and process of lipid with human. The aim for this study is to asses the effect of Moringa oleifera to cholesterol levels on diet induced obesity zebrafish. This experimental study is done with post test only control group design. Subjected zebrafish is adult female within 5 months post fertilisation age. Zebrafish population is randomly assigned to five groups of different treatments: control group negative (as a non-DIO), control group positive (DIO), and DIO group treated with Moringa oleifera leaves extract with a dose of 0,56 ppm, 1,12 ppm, and 2,24 ppm. Variables of total cholesterol, HDL level, and LDL/VLDL level is then measured from extracted visceral fat of zebrafish with Cholesterol Assay. The result shows that a significant lipid profile increase could be observed in DIO group. Moringa oleifera extract given to diet induced zebrafish shows decreased total cholesterol (p=0.002), increased HDL (p=0.00), and lower LDL/VLDL (p=0.00) than DIO group. However increases of dose to 1,12 ppm and 2,24 ppm does not show significant lipid profile improvement.
