Abstract

Traumatic Brain Injury (TBI) merupakan perubahan dalam fungsi otak yang bermanifestai sebagai kebinggungan, tingkat kesadaran yang berubah, kejang, koma, dan defisit motoris neurologis diakibatkan tekanan benda tumpul atau penetrasi benda tajam masuk dalam kepala. Penelitian ini bertujuan untuk mengetahui efek terapi menggunakan ekstrak kulit manggis (Garcinia mangostana L.) terhadap penurunan protease plasma darah dan peningkatan ekspresi IL-10 pada otak tikus putih (Rattus norvegicus) model TBI. Hewan coba yang dipakai yaitu tikus putih (Rattus norvegicus) jantan berumur 2-3 bulan yang dibagi menjadi empat kelompok perlakuan yakni kelompok tikus kontrol, kelompok tikus TBI, kelompok terapi ekstrak kulit manggis dan kelompok terapi minocycline. Aktivitas protease dihitung menggunakan metode spektrofotometri. Ekspresi IL-10 dianalisis dengan menggunakan metode imunohistokimia. Hasil penelitian menunjukkan bahwa terapi ekstrak kulit manggis secara signifikan (p < 0,01) menurunkan aktifitas protease sebesar 37,86 %. Terapi ekstrak kulit manggis secara signifikan (p < 0,01) mampu meningkatkan ekspresi IL-10 sebesar 35,94 %. Dapat disimpulkan bahwa terapi ekstrak kulit manggis mampu menurunkan aktivit

English Abstract

Traumatic Brain Injury (TBI) is a change in the function of the brain that manifests as a turnaround, the level of altered consciousness, convulsions, coma, and neurological motor deficit caused by blunt object pressure or penetration of sharp objects inside the head. The aim of this study was to investigate the effect of therapy using mangosteen skin extract (Garcinia mangostana L.) on the decrease of blood plasma protease and increased expression of IL-10 on brain’s TBI rats (Rattus norvegicus). The experimental animals were male rats (Rattus norvegicus) aged 2-3 months divided into four treatment groups ie control group of rats, group of TBI rats, mangosteen peel extract therapy group and minocycline therapy group. Protease activity was calculated using spectrophotometric method. The expression of IL-10 was analyzed using immunohistochemical methods. The results showed that treatment of mangosteen peel extract significantly (p < 0,01) decreased the protease activity up to 37.86%. Treatment of mangosteen peel extract significantly (p < 0,01) was able to increase IL-10 expression up to 35.94%. It can be concluded that mangosteen peel extract therapy can decrease protease activity and increase IL-10 expression in TBI rats.
