Abstract
Biji buncis (Parkia timoriana) merupakan makanan ringan yang populer di kalangan masyarakat Jawa, kelompok etnis terbesar di Indonesia. Mungkin saja hidrolisatnya termasuk peptida aktif karena mengandung 30% protein. Sejumlah penelitian menyarankan efek antidiabetes dan antihipertensi dari genusnya. Sayangnya tidak ada penelitian sebelumnya tentang penghambat DPP-IV dan ACE, khususnya pada Parkia timoriana. Oleh karena itu, tujuan dari penelitian ini adalah untuk menganalisis protein kandungan hidrolisat biji pohon (TBP) untuk DPP-IV dan ACE-inhibiting peptida. TBP dicerna oleh trypsin, thermolysin, chymotrypsin, pepsin, dan Enzim SGI untuk menghasilkan peptida bioaktif. Pencernaan SGI menghasilkan sebagian besar hidrolisis (DH) sebesar 13,7%. Setiap hidrolisat diuji untuk DPP-IV dan penghambatan ACE. Hidrolisat SGI paling efektif menghambat DPP-IV dan KARTU AS. Hidrolisat SGI menjadi sasaran bioassay-guided berurutan fraksinasi dengan pertukaran kation kuat (SCX) dan kinerja tinggi fase terbalik kromatografi cair untuk mengidentifikasi peptida penghambatan DPP-IV (RP-HPLC). Fraksi SCX (F2) terbaik dipisahkan dengan RP-HPLC untuk mendapatkan fraksi H11 yang memiliki IC50 sebesar 0,9499 0,05 mg/mL. Menggunakan sintetis peptida, tiga penghambat DPP-IV yang kuat diidentifikasi pada prinsipal puncak. Urutannya meliputi FVLNAPY (FY7) (IC50 > 2000 M), DTLGLGVSNL (DL-10) dan APLGPF (AF6) (IC50 396 M), dengan APLGPF (AF6) menjadi penghambat DPP-IV yang paling kuat. Pada IC50 72,27 M, FVLNSPY (FY7) adalah penghambat ACE yang paling kuat. Mereka tahan terhadap SGI eznyme. Docking molekul memvalidasi kesimpulan ini. Peptida aktif diproduksi dari hidrolisat TBP dapat digunakan untuk mengobati diabetes dan hipertensi.
English Abstract
Tree bean (Parkia timoriana) seeds are a popular snack among the Javanese, the largest ethnic group in Indonesia. It is possible that its hydrolysate includes active peptides because it contains 30% protein. Numerous studies suggest antidiabetic and antihypertensive effects of its genus. Unfortunately, there is no prior research on DPP-IV and ACE inhibitors, particularly on Parkia timoriana. Therefore, the purpose of this research was to analyze the protein content of tree beans (TBP) hydrolysates for DPP-IV and ACE-inhibiting peptides. TBP was digested by trypsin, thermolysin, chymotrypsin, pepsin, and SGI enzymes to produce the bioactive peptide. SGI digestion resulted in the most hydrolysis (DH) at 13.7%. Each hydrolysate was tested for DPP-IV and ACE inhibition. The SGI hydrolysate most effectively inhibited DPP-IV and ACE. SGI hydrolysate was subjected to sequential bioassay-guided fractionation with strong cation exchange (SCX) and reversed-phase highperformance liquid chromatography to identify DPP-IV inhibitory peptides (RP-HPLC). The best fraction of SCX (F2) was separated by RP-HPLC to get fraction H11, which had an IC50 of 0.9499 0.05 mg/mL. Using synthetic peptides, three powerful DPP-IV inhibitors were identified in the principal peaks. Its sequences include FVLNAPY (FY7) (IC50 > 2000 M), DTLGLGVSNL (DL-10) and APLGPF (AF6) (IC50 396 M), with APLGPF (AF6) being the most potent DPP-IV inhibitor. At an IC50 of 72.27 M, FVLNSPY (FY7) was the most potent ACE inhibitor. They are resistant to SGI eznyme. Molecular docking validated this conclusion. Active peptides produced from TBP hydrolysate may be used to treat diabetes and hypertension.
