Abstract
Meningkatnya populasi penduduk setiap tahunnya menyebabkan kebutuhan pangan semakin meningkat. Sehingga harus ditunjang dengan produktivitas yang tinggi terkait dengan beras. Tetapi terdapat beberapa kendala yang menyebabkan produktivitas padi menurun, salah satunya yaitu Hawar Daun Bakteri (HDB) yang disebabkan oleh bakteri patogen Xanthomonas oryzae pv. oryzae (Xoo). Salah satu pengendalian yang dilakukan yaitu menggunakan pestisida kimia. Sehingga diperlukan sebuah bahan aktif yang mempunyai potensi untuk mengendalikan bakteri Xoo. Salah satunya yaitu menggunakan bahan aktif Mankozeb 80%. Penggunaan bahan aktif mankozeb juga harus disesuai dengan dosis yang digunakan serta harus mengetahui efek pasca pengaplikasian bahan aktif mankozeb salah satunya terkait keberadaan bakteri antagonis. Tujuan penelitian ini yaitu mengetahui rekomendasi konsentrasi penggunaan bahan aktif mankozeb 80% terhadap penekanan perkembangan bakteri Xoo. Selain itu mengetahui toleransi bakteri filosfer pada daun tanaman padi terhadap bahan aktif mankozeb 80%, serta mengetahui potensi bakteri filosfer. Penelitian dilakukan pada bulan Oktober 2022 sampai Januari 2023. Pengambilan sampel dilakukan di Kecamatan Junrejo Kota Batu, dan untuk penelitian skala in vitro dilakukan di Laboratorium Penyakit Tumbuhan 1 Departemen Hama dan Penyakit Tumbuhan Universitas Brawijaya dan Laboratorium Bakteri PT Petrokimia Kayaku, Gresik. Metode yang dilakukan dalam penelitian daya hambat mankozeb terhadap bakteri Xoo menggunakan RAL dengan 5 perlakuan dan 4 ulangan. Perlakuan menggunakan dosis 1,5 gr/l, 2,25 gr/l, 3 gr/l dan 6 gr/l serta kontrol. Selain itu dalam penelitian uji daya hambat bakteri filosfer terhadap Xoo menggunakan RAL dengan 5 perlakuan dan 4 Ulangan. Bakteri yang dilakukan uji yaitu bakteri TM 4, TM 12, TM 14, dan TM 16. Jika terdapat perbedaan nyata maka dilakukan uji lanjut DUNCAN dengan taraf kesalahan 5%. Dalam melakukan seleksi bakteri antagonis dilakukan uji toleransi mankozeb, kemudian dilakukan uji hipersensitif,uji patogenitas, dan kemudian dilakukan identifikasi. Berdasarkan hasil penelitian yang telah dilakukan maka didapatkan hasil bahwa dosis mankozeb yang efektif mengendalikan bakteri Xoo yaitu dengan dosis 2.25 gr/l. Kemudian terdapat bakteri filosfer yang masih bisa toleran bahan aktif mankozeb 80%. Selain itu juga didapatkan bakteri filosfer yang bukan pathogen sejumlah 14 isolate bakteri non pathogen isolate tersebut yaitu TM 3, TM 4, TM 5, TM 6, TM 7, TM 8, TM 9, TM 11, TM 12, TM 13, TM 15, TM 16, dan TM 17. Dari 14 isolate bakteri terdapat 4 isolate bakteri yang mempunyai potensi bersifat antagonis yaitu isolate bakteri TM 4, TM 12, TM 14, dan TM 16. Bakteri antagonis Sebagian besar dari gram negatif. Dari genus Corynebacterium spp. dan Bacillus spp.
English Abstract
The increasing people every year causes the need for food to increase. So it must be supported by high productivity associated with rice. But there are several obstacles that cause rice productivity to decrease.The plant disease that causes the most losses is Bacterial Leaf Blight (HDB) caused by the pathogenic bacterium Xanthomonas oryzae pv. oryzae (Xoo). One of the controls carried out is using chemical pesticides, but a few bactericidal can be used to control Xoo bacteria. So we need an active ingredient that has the potential to control the Xoo bacteria. One of them is using the active ingredient Mancozeb 80%. The use of the active ingredient mancozeb must also be in accordance with the dosage used and must know the effects after application of the active ingredient mancozeb, one of which is related to the presence of antagonistic bacteria. The purpose of this study was to find out the concentration recommendations for using the active ingredient mancozeb 80% to suppress the development of Xoo bacteria. In addition to knowing the tolerance of phylospheric bacteria in the leaves of rice plants to the active ingredient mancozeb 80%, as well as knowing the potential of philospheric bacteria. The research was conducted from October 2022 to January 2023. Sampling was carried out in Junrejo District, Batu City, and for in vitro scale research it was carried out at the Plant Disease Laboratory 1, Department of Plant Pests and Diseases, Brawijaya University and the Bacteria Laboratory of PT Petrokimia Kayaku, Gresik. The method used in the study of the inhibition of mancozeb against Xoo bacteria used RAL with 5 treatments and 4 replications. The treatment used doses of 1.5 gr/l, 2.25 gr/l, 3 gr/l and 6 gr/l as well as controls. In addition, in the study of the inhibition of phloosphere bacteria against Xoo, RAL was used with 5 treatments and 4 replications. The bacteria tested were TM 4, TM 12, TM 14, and TM 16. If there is a significant difference, then the DUNCAN test is carried out with an error level of 5%. In selecting antagonistic bacteria, a mancozeb tolerance test was carried out, then a hypersensitivity test, pathogenicity test was carried out, and then identification was carried out. Based on the results of the research that has been done, it was found that the effective dose of mancozeb in controlling Xoo bacteria was at a dose of 2.25 gr/l. Then there are philospheric bacteria that can still tolerate 80% of the active ingredient mancozeb. In addition, 14 non-pathogenic bacterial isolates were also found, namely TM 3, TM 4, TM 5, TM 6, TM 7, TM 8, TM 9, TM 11, TM 12, TM 13, TM 15, TM 16, and TM 17. Of the 14 bacterial isolates, there were 4 bacterial isolates that had antagonistic potential, namely bacterial isolates TM 4, TM 12, TM 14, and TM 16. Antagonistic bacteria Most of them are gram negatif. From the genus Corynebacterium spp. and Bacillus spp.
