Abstract

Jeruk merupakan tanaman buah yang memiliki tingkat produksi tinggi di Indonesia. Salah satu penyebab penurunan produktivitas tanaman jeruk adalah serangan penyakit kanker jeruk yang disebabkan oleh bakteri Xanthomonas axonopodis pv. citri (Xac). Pengendalian yang dilakukan oleh petani adalah menggunakan bahan kimia berupa antibiotik seperti streptomisin. Pengendalian hayati merupakan salah satu alternatif yang dapat dilakukan untuk mengendalikan penyakit kanker jeruk. Salah satu pengendalian hayati adalah dengan menggunakan bakteri yang bersifat antagonis terhadap bakteri Xac. Bakteri filosfer yang bersifat antagonis dapat ditemukan pada tanaman jeruk. Penelitian ini dilakukan untuk mengetahui potensi antagonis bakteri filosfer terhadap bakteri Xac dan mengetahui genus dari bakteri. Kegiatan penelitian dilakukan di Laboratorium Terpadu Balai Penelitian Tanaman Jeruk dan Buah Subtropika dari bulan agustus hingga desember 2022. Tahapan penelitian ini meliputi: isolasi bakteri filosfer dari daun tanaman jeruk, purifikasi bakteri filosfer, uji patogenisitas bakteri Xac pada tanaman jeruk, perbanyakan bakteri Xac pada media padat, perbanyakakan bakteri filosfer dan bakteri Xac pada media cair, seleksi potensi antagonis bakteri filosfer, uji efektivitas bakteri filosfer pada daun jeruk, dan identifikasi bakteri filosfer. Penelitian ini disusun dengan menggunakan metode Rancangan Acak Lengkap (RAL). Uji efektivitas dilakukan menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 ulangan. Hasil isolasi bakteri filosfer dari tanaman jeruk diperoleh 37 isolat bakteri filosfer. Pada tahap seleksi potensi bakteri bersifat antagonis diambil 3 isolat dengan zona hambat tertinggi. Bakteri dilakukan identifikasi pada tingkat genus dan didapatkan isolat F6 dan F18 dari genus Erwinia dan isolat F26 dari genus Burkholderia. Bakteri filosfer dilakukan uji efektivitas dibandingkan dengan perlakuan bakterisida dan perlakuan kontrol dan didapatkan berbeda nyata dengan perlakuan kontrol.

English Abstract

Orange is a fruit plant that has a high production level in Indonesia. One of the causes of decreased productivity of citrus plants is the attack of citrus canker disease caused by the bacterium Xanthomonas axonopodis pv. citri (Xac). The control carried out by farmers is using chemicals in the form of antibiotics such as streptomycin. Biological control is an alternative that can be done to control citrus cancer. One of the biological control is to use bacteria that are antagonistic to Xac bacteria. Philospheric bacteria that are antagonistic can be found in citrus plants. This research was conducted to determine the antagonistic potential of phyllospheric bacteria against Xac bacteria and to determine the genus of the bacteria. Research activities were carried out at the Integrated Laboratory of the Indonesian Citrus and Subtropical Fruit Research Institute from August to December 2022. The stages of this research included: isolation of phylospheric bacteria from citrus leaves, purification of philospheric bacteria, test of pathogenicity of Xac bacteria on citrus plants, multiplication of Xac bacteria and phylospheric bacteria in liquid media, selection of antagonist potential of philospheric bacteria, testing the effectiveness of philospheric bacteria on lime leaves, and identification of philospheric bacteria. This study was arranged using a completely randomized design (CRD). The effectiveness test was carried out using a completely randomized design (CRD) with 5 treatments and 3 repetition. The results of the isolation of phylospheric bacteria from citrus plants obtained 37 isolates of phylospheric bacteria. At the selection stage for the potential of antagonistic bacteria, 3 isolates with the highest inhibition zone were taken. Bacteria were identified at the genus level and isolates F6 and F18 from the genus Erwinia and isolates F26 from the genus Burkholderia were obtained. Philospheric bacteria were tested for effectiveness compared to the bactericidal treatment and the control treatment and found to be significantly different from the control treatment.
