Abstract
Salah satu penyebab penurunan hasil panen pada tanaman terung ungu adalah adanya gangguan OPT (Organisme Pengganggu Tanaman). OPT yang menyerang tanaman terung ungu adalah Cucumber Mosaic Virus. Penendalian yang dapat digunakan adalah dengan menginduksi ketahanan sistemik. Ketahanan sistemik tanaman dapat diaktifkan dengan menginduksi gen-gen ketahanan yang terdapat di dalam tanaman menggunakan kitosan. Kitosan adalah produk alami yang berasal dari ekstrak cangkang udang ataupun cangkang kepiting. Manfaat kitosan sangat banyak, salah satunya yaitu dibidang pertanian untuk menghambat virus. Pengaruh berat molekul kitosan terhadap kemampuannya menekan infeksi sistemik bean mild mosaic virus pada tanaman kacang (Phaseolus vulgaris L.) dipelajari Tujuan penelitian ini adalah untuk mengetahui kemampuan kitosan dalam menekan infeksi Cucumber Mosaic Virus (CMV) dan pengaruh berbagai frekuensi pemberian kitosan terhadap masa inkubasi, intensitas serangan, tinggi tanaman, jumlah daun, dan kadar klorofil tanaman terung ungu. Penelitian di Laboratorium Toksikologi Pestisida dan Rumah Kaca Jurusan Hama dan Penyakit Tumbuhan serta di Laboratorium Bioteknologi Departemen Agronomi Pertanian Fakultas Pertanian Universitas Brawijaya, Malang pada bulan Juli hingga bulan Oktober 2022. Alat yang digunakan dalam penelitian ini adalah adalah gelas ukur (vol. 100 ml), magnetic stirrer, spektrotometer uv vis, tabung reaksi, cuvet, pipet mikro, cawan petri, timbangan analitik, kain kasa, tisu, botol semprot, mortar, pistil, polibag 35 x 35 cm, cangkul, botol kaca (5ml), kertas label, suntikan, isolasi, gunting dan kamera. Bahan yang digunakan dalam penelitian ini adalah kitosan, tanah, aram sekam, pupuk NPK, kompos, benih terung ungu, inokulum CMV, tanaman indikator Gomphorena globosa dan Vignia unguiculata, karborundum 600 mesh, aquades, buffer phospat pH 7, alkohol 70%, asam asetat, NaOH, aseton 80% dan formalin 4%. Percobaan dilakukan menggunakan metode Rancangan Acak Lengkap (RAL) terdiri atas inokulasi CMV, Inokulasi CMV dan 1 kali aplikasi kitosan, Inokulasi CMV dan 2 kali aplikasi kitosan, Inokulasi CMV dan 3 kali aplikasi kitosan, Inokulasi CMV dan 4 kali aplikasi kitosan, kontrol. Variabel pengamatan berupa masa inkubasi, intensitas serangan penyakit, tinggi tanaman, jumlah daun, dan total kadar klorofil. Selanjutnya, dilakukan analisis ragam (ANOVA) taraf 5%. Jika perlakuan berpengaruh nyata terhadap hasil pengamatan, maka dilakukan uji lanjut dengan uji beda nyata jujur (BNJ) pada taraf 5%. Dari hasil penelitian dapat diketahui bahwa kitosan yang diaplikasikan dengan frekuensi yang berbeda dapat mempengaruhi intensitas dari serangan CMV, tinggi tanaman, jumlah daun, dan kandungan klorofil pada tanaman mentimun serta dapat memperlambat masa inkubasi. Perlakuan yang mampu memberikan tekanan untuk meningkatkan ketahanan tanaman adalah aplikasi kitosan dengan frekuensi empat kali dibandingkan dengan perlakuan lainnya dan memiliki hasil yang tidak berbeda jauh dengan perlakuan kontrol.
English Abstract
One of the causes of decreased yields on purple eggplant plants is the presence of Pest and Disease Organism disturbances. One of the disease that attack the purple eggplant is the Cucumber Mosaic Virus. Method that can be used to control plant pathogens is by induced plants resistance. Plant systemic resistance can be activated by inducing resistance genes present in plants by used chitosan. Chitosan is a natural product derived from the extract of shrimp shells or crab shells. The benefits of chitosan are numerous, one of which is in the agricultural sector to inhibit viruses. The effect of the molecular weight of chitosan on its ability to suppress systemic infection of bean mild mosaic virus in bean (Phaseolus vulgaris L.) plants was studied. The purpose of this study was to determine the ability of chitosan to suppress Cucumber Mosaic Virus (CMV) infection and the effect of various frequencies of chitosan administration on the incubation period, attack intensity, plant height, number of leaves, and chlorophyll content of purple eggplant. This research was conducted at the Pesticide Toxicology and Greenhouse Laboratory, Department of Plant Pests and Diseases and at the Biotechnology Laboratory, Department of Agricultural Agronomy, Faculty of Agriculture, University of Brawijaya, Malang from July to October 2022. The tools used in this study was a measuring cup (vol. 100 ml), magnetic stirrer, uv vis spectrotometer, test tube, cuvette, micro pipette, petri dish, analytical balance, gauze, tissue, spray bottle, mortar, pistil, polybag 35 x 35 cm, hoe, glass bottle (5 ml), label paper, syringe, tape, scissors and camera. The materials used in this study were chitosan, soil, rice husk, NPK fertilizer, compost, purple eggplant seed, CMV inoculum, indicator plants Gomphorena globosa and Vignia unguiculata, carborundum 600 mesh, distilled water, phosphate buffer pH 7, alcohol, acetic acid, NaOH, 80% acetone and 4% formalin. The experiment was carried out using a completely randomized design (CRD) method in the form of treatment consisting of CMV inoculation, CMV inoculation and 1 chitosan application, CMV inoculation and 2 chitosan applications, CMV inoculation and 3 chitosan applications, CMV inoculation and 4 chitosan applications, control . The observed variables were incubation period, intensity of disease attack, plant height, number of leaves, and total chlorophyll content. Next, an analysis of variance (ANOVA) was performed at the 5% level. If the treatment has a significant effect on the results of the observations, then a further test is carried out with BNJ at the 5% level. From the results of the research conducted, it can be seen that chitosan applied with different frequencies can affect the intensity of CMV attack, plant height, number of leaves, and chlorophyll content in cucumber plants and can slow down the incubation period. The best treatment in this research was shown in the the 4 times application treatment was only able to exert pressure to increase plant resistance to CMV
