Abstract
Tomat adalah tanaman sayuran yang peminatnya tinggi. Faktor tingginya peminat tomat karena berperan sebagai sayuran, jus, campuran bumbu masak, bahan baku industri dan mampu menyembuhkan berbagai penyakit. Permintaan tomat dari tahun ke tahun terus meningkat namun petani mengalami kendala dalam meningkatkan produksi tomat, salah satu masalah penting yang dihadapi adalah penyakit layu bakteri Ralstonia solanacearum. Serangan berat mampu menyebabkan tanaman yang dibudiayakan mati hingga gagal panen. Oleh karena itu, perlu dilakukan pengendalian penyakit layu bakteri untuk menekan intensitas serangan penyakit. Pengendalian dapat dilakukan dengan memperkuat jaringan tanaman dengan mengaplikasikan pupuk kandang dan agens hayati Trichoderma harzianum. Pengujian ini bertujuan untuk mengetahui pengaruh pengaplikasian pupuk kandang dan agens hayati terhadap intensitas serangan penyakit layu bakteri dan pertumbuhan tanaman tomat. Kegiatan penelitian dilaksanakan di Desa Sumberejo, Kecamatan Geger, Kabupaten Madiun, Provinsi Jawa Timur. Laboratorium Penyakit Tumbuhan Jurusan Hama dan Penyakit Tumbuhan, Universitas Brawijaya. Laboratorium tanah Balai Pengkajian Teknologi Pertanian Jawa Timur. Alat dan bahan yang digunakan adalah tray, polybag, drum, cawan petri, autoclave, LAF, shaker, kompor, kertas label, tissue, alat tulis, kamera, tanah, pupuk kandang sapi, pupuk kandang ayam, isolat Trichoderma harzianum, benih tomat, aquades, alkohol 70 %, media NA, media NB, media PDA, media EKG, tanaman tomat terserang penyakit layu bakteri. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), menggunakan 6 perlakuan dan 5 ualangan. Analisis data dilakukan menggunakan uji F (ANOVA) dengan selang kepercayaan 5%. Jika uji F menghasilkan data yang berbeda nyata maka dilanjutkan dengan Uji BNJ dengan taraf 5 %. Pelaksanaan Penelitian meliputi persiapan isolasi, pembuatan media, isolasi Ralstonia solanacearum, analisis kandungan pupuk kandang, uji mutu agens hayati, penyemaian, sterilisasi media tanam, penanaman, inokulasi Ralstonia solanacearum dan aplikasi agens hayati, serta pemeliharaan. Parameter pengamatan yang diamati dalam penelitian ini adalah intensitas serangan penyakit, tinggi tanaman, dan jumlah daun. Berdasarkan hasil pengamatan, dapat disimpulkan bahwa pengaplikasian pupuk kandang yang dikombinasikan dengan agens hayati Trichoderma harzianum mampu mengendalikan intensitas penyakit layu bakteri Ralstonia solanacearum dan dapat mengoptimalkan pertumbuhan tanaman tomat. Berdasarkan hasil penelitian, disarankan melakukukan pengamatan hingga panen, dikarenakan salah satu parameter keberhasilan dalam pengaplikasian pupuk kandang dan Trichoderma harzianum adalah produksi tanaman tomat.
English Abstract
Tomato is vegetable that is in high demand. The reason for the high demand in tomato is that they function as vegetables, juices, a mixture of cooking spices, and industrial raw materials and can cure various diseases. The demand for tomato from year to year continues to increase but farmers experience problems in increasing tomato production, one of the important problems faced is the bacterial wilt disease Ralstonia solanacearum. Weight attacks can cause the cultivated plants to die until harvest failure. Therefore, it is necessary to control bacterial wilt disease to reduce the intensity of disease attacks. Control can be done by strengthening plant tissue by applying manure and biological agents Trichoderma harzianum. This research aims to determine the effect of the application of manure and biological agents on the intensity of bacterial wilt disease and tomato plant growth. The research activity was carried out in Sumberejo, Geger, Madiun, and East Java. Plant Diseases Laboratory, Department of Plant Pests and Diseases, Universitas Brawijaya. Soil laboratory of the Balai Pengkajian Teknologi Pertanian Jawa Timur. Tools and materials used are trays, polybag, drum, petri dish, autoclave, LAF, shaker, stove, label, tissue, stationery, camera, soil, cow manure, chicken manure, isolate Trichoderma harzianum, tomato seeds, equates, alcohol 70 %, NA media, NB media, PDA media, EKG media, tomato plants were attacked by bacterial wilt disease. This research used a completely randomized design (CRD), using 6 treatments and 5 replications. Data analysis was performed using the F test (ANOVA) with a 5 % confidence interval. If the F test produces significantly different data, then it is continued with the BNJ test with a level of 5%. The implementation of the research includes preparation of isolation, manufacture of media, isolation of Ralstonia solanacearum, analysis of manure content, a quality test of biological agents, seeding, sterilization of planting media, planting, inoculation of Ralstonia solanacearum, and application of biological agents, and maintenance. Parameters observed in this study were the intensity of disease attack, plant height, and several leaves. Based on the observations, it can be concluded that the application of manure combined with the biological agent Trichoderma harzianum is able to control the intensity of the bacterial wilt disease Ralstonia solanacearum and can optimize the growth of tomato plants. Based on the results of the study, it is recommended to observe until harvest, because one of the parameters of success is the application of manure and Trichoderma harzianum in the production of tomato plants.
