Abstract
Dampak negatif dari penggunaan insektisida kimia bagi makhluk hidup dan lingkungan telah menjadikan jamur patogen serangga (JPS) Beauveria bassiana sebagai alternatif pengendalian hama. Pemanfaatan JPS B. bassiana bergantung pada produksi massal yang mudah, murah, dan infentif. Di Indonesia, limbah pertanian yang dihasilkan setiap tahun cukup tinggi, sehingga penggunaan media limbah seperti sekam padi, ampas teh, bekatul padi, dan dedak gandum dapat menjadi media produksi JPS B. bassiana. Untuk meningkatkan patogenisitas dan produksi konidia maka diperlukan teknik perbanyakan tertentu seperti penambahan tepung cengkerik. Pengendalian hayati menggunakan JPS B. bassiana ditentukan oleh isolat dengan patogenisitas tinggi. Tingkat patogenisitas isolat tidak hanya dipengaruhi oleh genetik isolat, tetapi juga bergantung pada jenis media produksi. Jenis media produksi menentukan tingkat pertumbuhan JPS B. bassiana. Penelitian ini dilaksanakan mulai dari bulan April 2022 sampai Juli 2022 di Laboratorium Pengendalian Hayati Fakultas Pertanian Universitas Brawijaya, Malang. Penelitian dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL) dengan 9 perlakuan (sekam padi, ampas teh, bekatul padi, dedak gandum, sekam padi diperkaya tepung cengkerik, ampas teh diperkaya tepung cengkerik, bekatul padi diperkaya tepung cengkerik, dedak gandum diperkaya tepung cengkerik) dan kontrol (PDA). Setiap perlakuan diulang sebanyak 3 kali. Pengamatan dilakukan pada pertumbuhan diameter koloni, produksi konidia, mortalitas, dan Lethal Time (LT50) larva Spodoptera litura instar tiga. Berdasarkan hasil uji ANOVA, diketahui bahwa bekatul padi diperkaya tepung cengkerik (6,60 cm) dan dedak gandum diperkaya tepung cengkerik (7,43 cm). menjadi media yang paling sesuai untuk pertumbuhan diameter koloni isolat B. bassiana. Produksi konidia dan mortalitas tertinggi dihasilkan pada bekatul padi diperkaya tepung cengkerik dengan kerapatan konidia 30,23x107 konidia/ml pada 14 HSI, viabilitas 83,38%, dan mortalitas 100% pada 7 HSA. Sementara nilai Lethal Time (LT50) dari setiap perlakuan menunjukkan bekatul padi diperkaya tepung cengkerik menjadi media limbah yang menyebabkan kematian 50% larva S. litura tercepat, yaitu selama 3,25 hari.
English Abstract
The negative impact of the use of chemical insecticides on organisms and the environment has made insect pathogenic fungi (JPS) Beauveria bassiana an alternative pest control. Utilization of JPS relies on easy, inexpensive, and inventive mass production. In Indonesia, agricultural waste generated annually is quite high, so the use of waste media such as rice husks, tea leaf waste, rice pollard, and wheat bran can be used as new JPS production media. To increase the virulence and conidia production, certain propagation techniques are needed, such as the addition of cricket powder. Biological control using JPS B. bassiana was determined by isolates with high pathogenicity. The level of pathogenicity of isolates is not only influenced by the genetics of the isolates, but also depends on the type of production medium. The type of production media determined the growth rate of JPS B. bassiana. This research was conducted from April 2022 to July 2022 at the Laboratory of Biological Control, Faculty of Agriculture, University of Brawijaya, Malang. The study was conducted using a completely randomized design (CRD) with 9 treatments (rice husk, tea leaf waste, rice pollard, wheat bran, rice husk enriched with cricket powder, tea leaf waste enriched with cricket powder, rice pollard enriched with cricket powder, wheat bran enriched with cricket powder) and control. Each treatment was repeated 3 times. Observations were made on the growth of colony diameter, conidia production, viability, mortality, and Lethal Time (LT50) of third instar Spodoptera litura larvae. Based on the ANOVA test results, it was found that rice pollard enriched with cricket powder (6.60 cm) and wheat bran enriched with cricket powder (7.43 cm) were the most suitable media for the growth of colony diameter. The highest conidia production, viability, and mortality were produced by rice pollard enriched with cricket powder with conidia density of 30.23x107 conidia/ml at 14 DAP, viability 83.38%, and 100% mortality at 7 DAA. Meanwhile, the Lethal Time (LT50) value of each treatment showed that rice pollard enriched with cricket powder was the best waste medium because it caused the fastest 50% mortality of S. litura larvae, which was 3.25 days.
