Abstract
Tembakau (Nicotiana tabaccum L.) merupakan komoditas pertanian yang Di Indonesia. Produksi tembakau mencapai 261,40 ribu ton pada tahun 2020. Tembakau di Indonesia selama ini dimanfaatkan untuk pembuatan rokok. Namun, konsumsi rokok yang berlebihan dapat mengakibatkan terganggunya kesehatan tubuh karena adanya kandungan nikotin dalam tembakau. Daun tembakau memiliki kandungan alkaloid, saponin, flavonoid, dan polifenol. Kandungan flavonoid dan alkaloid bermanfaat sebagai antibakteri, antijamur, dan anti serangga. Penelitian ini bertujuan untuk melakukan optimasi proses ekstraksi daun tembakau sebagai sediaan antibakteri. Faktor yang digunakan yaitu rasio bahan : pelarut (b/v) dengan level faktor 1:3; 1:5; 1:7 (b/v) dan waktu ekstraksi dengan level 4 menit, 6 menit, dan 8 menit. Metode ekstraksi yang digunakan adalah microwave assisted extraction (MAE), pengolahan data menggunakan metode Respon Surface Methodology (RSM). Hasil penelitian ini menunjukkan bahwa faktor rasio bahan : pelarut dan waktu ekstraksi berpengaruh nyata terhadap nilai total flavonoid, daya hambat ekstrak terhadap bakteri S. mutans, dan rendemen ekstrak. Namun, tidak berpengaruh nyata terhadap pH ekstrak. nilai total flavonoid, daya hambat, dan pH tertinggi didapatkan dari perlakuan rasio 1:2 (b/v) dan waktu 6 menit, nilainya berturut-turut yaitu 524,67 mgEQ/g; 22,4 mm; dan 6,6. Adapun nilai rendemen tertinggi didapatkan dari perlakuan rasio 1:5 (b/v) dan waktu 6 menit yaitu 4,59%. Hasil solusi optimal proses ekstraksi daun tembakau adalah perlakuan 1:2,6 (b/v) dan waktu 7 menit dengan hasil total flavonoid, daya hambat ekstrak, pH, dan rendemen berturut-turut 428,4 mgEQ/g; 20,7 mm; 5,8; dan 3,1%.
English Abstract
Tobacco (Nicotiana tabaccum L.) is an agricultural commodity whose tobacco production in Indonesia reached 261.40 thousand tons in 2020. Tobacco in Indonesia has so far been used to make cigarettes. However, excessive consumption of cigarettes can cause health problems due to the nicotine in tobacco. Tobacco leaves contain alkaloids, saponins, flavonoids, and polyphenols. The content of flavonoids and alkaloids is useful as an antibacterial, antifungal, and anti-insect. This study aims to optimize the extraction process of tobacco leaves as an antibacterial preparation. The factors used are the ratio of material: solvent (w/v) and extraction time. The extraction method used is microwave assisted extraction (MAE), data processing using the response surface methodology (RSM). The results of this study showed that the ratio of ingredients: solvent and extraction time significantly affected the total flavonoid value, the inhibitory power of the extract against S. mutans bacteria, and the extract yield. However, it had no significant effect on the pH of the extract. The highest total flavonoid value, inhibitory power, and pH were obtained from the treatment ratio 1:2 (w/v) and the value for 6 minutes was 524.67 mgEQ/g; 22.4 mm; and 6.6. The highest yield value was obtained from the treatment ratio of 1:5 (w/v) and time of 6 minutes, namely 4.59%. The results of the optimal solution of the tobacco leaf extraction process were 1:2,6 (w/v) treatment and 7 minutes with total flavonoids, extract inhibitory power, pH, and yield 428.4 mgEQ/g; 20.7 mm; 5.8; and 3.1% respectively.
