Abstract

Bakteri endofit adalah bakteri saprofit yang hidup dan bersimbiosis dengan jaringan tanaman tanpa menimbulkan suatu gejala panyakit pada tanaman tersebut. Tujuan dari penelitian ini adalah untuk mengembangkan agen pengendalian hayati dari bakteri endofit penghasil enzim protease yang mampu mendegradasi protein penyusun komponen matrik ekstraseluler dari jamur Botryodiplodia theobromae penyebab penyakit diplodia pada tanaman Citrus spp. Metode: Metode penelitian yang digunakan terdiri dari beberapa tahapan meliputi pemurnian bakteri endofit, isolasi jamur diplodia, pengamatan morfologi bakteri endofit, uji pewarnaan gram, uji hipersensitif, uji antagonis, dan uji aktivitas enzim protease. Hasil: Hasil penelitian diperoleh bahwa sebanyak 9 isolat bakteri endofit yang mampu menghambat pertumbuhan jamur B. theobromae. Berdasarkan seleksi aktivitas enzim protease pada 10 isolat bakteri, diperoleh bahwa isolat AK3 memiliki indeks proteolitik terbesar yaitu sebesar 2,67 cm. Isolat AK3 merupakan bakteri Gram positif berbentuk batang, katalase positif, dan tidak bersifat patogen. Aktivitas enzim isolat AK3 memiliki waktu optimal pada hari ke-5 dengan nilai aktivitas enzim sebesar 0,1437 U/ml. Kesimpulan: Potensi bakteri endofit ini diharapkan mampu digunakan untuk kepentingan di berbagai industri.

English Abstract

Endophytic bacteria are saprophytic bacteria that live and are symbiotic with plant tissue without causing some symptoms in the plant. The purpose of this study is to develop a biological control agents from endophytic bacteria that produce protease enzymes that are able to degrade proteins making up the extracellular matrix components of the fungus Botryodiplodia theobromae that cause diplodia diseases in the Citrus spp. Method: The research methods used were several stages of purification of endophytic bacteria, isolation of diplodia fungi, observation of endophytic bacterial morphology, gram staining test, hypersensitive test, antagonistic test, test for protease enzyme activity. Results: The results obtained as many as 9 isolates of endophytic bacteria were able to fight the fungi B. theobromae. Based on the selection of protease enzyme activity in 10 bacterial isolates, AK3 isolates obtained the largest proteolytic index of 2.67 cm. AK3 isolates are Gram-positive bacteria in the form of bacill, are positive catalases, and are not pathogenic.. The enzyme activity of AK3 isolates had an optimal time on the 5th day with an enzyme activity value of 0.1437 U / ml. Conclusion: The potential of endophytic bacteria is expected to be used for interests in various industries.
