English Abstract
Herbal medicine is utilized for primary health care due to better social acceptability in the developing countries. One potential plant such as Entada Spiralis Ridley (synonym, Entada scheffleri) or locally identified as Sintok contains flavonoid, saponin, tannin, and glycoside. These compounds have anti-inflammatory, anti-fungal, anti-yeast and antibacterial activities. This research aims to identify the bioactive compounds and to determine the antimicrobial activity from crude extract E. spiralis. The crude extract was prepared by macerating the leaves with chloroform then fractionated by vacuum liquid chromatography with Dichloromethane : Hexane (1:9) and Dichloromethane : Methanol (9:1) solvent system. Evaluation of antimicrobial activity was further tested with Disk Diffusion Test. The highest zone inhibition against E.coli was obtained from fraction DCM:MeOH extract with diameter zone inhibition 12 mm. Chloroform crude extract was effective to kill S. aureus and C. albicans with zone inhibition of 14.50 and 10.33 mm, respectively. Chloroform crude extract was found to be more effective against pathogenic bacteria and fungi in microdilution assay, where the minimum inhibitory concentration were 1.563 mg/mL for E.coli and S.aureus, 0.781 mg/mL for C. albicans. Determination of bioactive compound was done by Thin Layer Chromatography (TLC), Total Phenolic Content (TPC) and Total Flavonoid Content (TFC). TPC of DCM:MeOH, Chloroform, and DCM:H were 50.56 ± 0.188, 51.913 ± 0.089 , 24.16 ± 0.175 mg GAE/g extract. Meanwhile the TFC were 16.96 ± 0.127, 17.35 ± 0.05, -0.27 ± 0.029 mg QE/g sample extract, respectively.
