Abstract
Pada tahun 2008, Indonesia melakukan ekspor bahan kosmetik berbasis organik dan natural, salah satunya berupa pewarna alami. Pewarna alami merupakan suatu zat yang berasal dari alam seperti tumbuh – tumbuhan. Salah satu tumbuhan yang dapat dimanfaatkan menjadi pewarna alami adalah buah naga. Buah naga merupakan jenis tanaman tropis yang memiliki kandungan antioksidan dan mikronutrien yang potensial. Pigmen betasianin merupakan pigmen yang paling banyak dijumpai di dalam buah ini. Pengambilan pigmen betasianin dilakukan dengan ekstraksi menggunakan Microwave Assisted Extraction (MAE). Tujuan dari penelitian ini yaitu untuk mengetahui volume pelarut dan waktu ekstraksi yang optimum terhadap kadar betasianin, aktivitas antioksidan, rendemen dan intensitas warna merah pada ekstrak buah naga dan potensi pengembangan pigmen betasianin sebagai lip balm. Metode optimasi dilakukan dengan menggunakan Response Surface Methodology (RSM) dengan rancangan Central Composite Design (CCD). Faktor pertama waktu ekstraksi terdiri dari 3 level (3, 5, 7) menit dan faktor kedua volume pelarut terdiri dari 3 level (125, 150, 175) ml. Respon penelitian ini antara lain kadar betasianin, aktivitas antioksidan (IC50), rendemen dan intensitas warna merah. Analisa data dilakukan menggunakan Design Expert 11 meliputi analisa pemilihan model ANOVA, dan penentuan kondisi optimum. Proses verifikasi kondisi optimum dilakukan sebanyak 3 kali perulangan. Hasil solusi optimum yang didapatkan adalah pada kombinasi waktu ekstraksi 3 menit dan volume pelarut 175 ml. Nilai pada kadar betasianin, IC50, rendemen dan intensitas warna merah hasil verifikasi masing – masing sebesar 247,317 ppm, 465,407 ppm, 2,28%, dan 6,3533. Tingkat akurasi hasil verifikasi pada respon kadar betasianin sebesar 85,84%, IC50 sebesar 81,9%, rendemen sebesar 99,8% dan intensitas warna merah sebesar 99,58%.
English Abstract
Indonesia was being the exporter of organic and natural cosmetic ingredients such as natural dyes in 2008. Natural dyes are the alternative colorant that comes from nature such as plants. Dragon fruits are one of the plants that can be used as natural dyes. This plant is categorized as tropical plants that contained antioxidants and micronutrients. Betacyanin is the most pigment contained in this plant. Mostly, extraction is a method used for releasing pigment from plants. There are so many methods of extraction, one of them is microwave assisted extraction methods. This research aims to know the optimum amount of solvents and extraction time on betacyanin content, antioxidants activities, yields, the red color intensity from dragon fruit extract, and potency of betacyanin pigments as a lip balm. Response Surface Methodology (RSM) method used for optimization process and Central Composite Design (CCD) with two factors used as experimental designed. The first factor was the extraction times with three-level (3, 5, 7) minutes and the second one was the amount of solvents with three-level (125, 150, 175) ml. The response of this experiment was betacyanin contents, antioxidant activities (IC50), yields, and red color intensity from dragon fruit extract. The data was analyzed by Design Expert 11 includes analysis of the ANOVA model and optimum conditions. The verification process of optimum conditions was repeated three times. The optimum solution results were a combination of 3 minutes extraction time and 175 ml of solvents. The results of the verification process on betacyanin contents, antioxidant activities (IC50), yields, and red color intensity were 247,317 ppm, 465,407 ppm, 2,28%, dan 6,3533. The accuracy of verification results on betacyanin contents, antioxidant activities (IC50), yields, and red color intensity were 85,84%, 81,9%, 99,8% 99,58%.
