Abstract
Jamur patogen Rhizoctonia sp. yang menyerang tanaman stroberi menjadi salah satu permasalahan yang sering terjadi dalam produksi buah tanaman stroberi, yang terjadi secara nasional dan internasional. Kontrol biologis dengan jamur endofit mampu menghasilkan hambatan pertumbuhan patogen melalui mekanisme kompetisi, mikoparasitisme, dan antibiosis. Penelitian ini bertujuan untuk mengetahui potensi jamur endofit dari tanaman stroberi sehat dalam menghambat pertumbuhan patogen Rhizoctonia sp.. Tahapan penelitian yang dilakukan adalah isolasi dan pemurnian jamur endofit dan patogen Rhizoctonia sp., identifikasi mikroskopis Rhizoctonia sp., pengujian antagonis dual culture pada lima isolat endofit terpilih (A.IV1.1, A.II9.1, D.III6.1, D.II9.1, dan B.III6.1), uji kualitatif enzim glukanase dan kitinase, serta identifikasi mikroskopis isolat endofit. Hasil penelitian menunjukkan bahwa perbedaan jenis isolat yang diisolasi berpengaruh nyata pada hasil penghambatan Rhizoctonia sp., dengan hasil daya hambat tertinggi oleh isolat A.IV1.1 (62,5%) pada masa inkubasi hari ke-5. Isolat B.III6.1, diketahui menghasilkan senyawa metabolit dengan terbentuknya zona kosong dan perubahan pigmen media disekitar koloni saat uji antagonis. Hasil uji kualitatif enzim glukanase dan kitinase dari kelima isolat menunjukkan hasil negatif. Identifikasi morfologi mikroskopis endofit menunjukkan genus kelima isolat, antara lain A.IV1.1 (Microclavia), D.III6.1 (Gliocladium), B.III6.1 (Periconia), A.II9.1 dan D.II9.1 (Aspergillus)
English Abstract
Fungal pathogen, Rhizoctonia sp. which infects strawberry plants is one of many problems that often occur in strawberry fruit production on a national and international scale. Biological control with endophytic fungi is able to inhibit growth of pathogens through various interaction mechanism, such as competition, mycoparasitism, and antibiosis. The purpose of this study is to determine the potential of endophytic fungi isolated from healthy strawberry, in inhibiting the growth of fungal pathogen Rhizoctonia sp.. The first steps of this study were isolation and purification of endophytic fungal isolates and fungal pathogen Rhizoctonia sp., microscopic morphological identification of Rhizoctonia sp., dual culture antagonist test on five selected isolates (A.IV1.1, A.II9.1, D.III61, D.II9.1, and B.III6.1), qualitative screening of glucanase and chitinase enzymes, and microscopic identification of endophytic fungal isolates. Results of this study showed that the types different in endophytic fungal isolates had a significant effect on the inhibition results of Rhizoctonia sp. growth, with highest inhibitory result by A.IV1.1 isolate (62,5%) on the 5th day of incubation. Endophytic fungal B.III6.1 isolate was known to produce metabolite compounds which indicated by the formation of empty zones and change in color of media pigment around colony during antagonist test. Qualitative screening test of glucanase and chitinase enzyme from five selected isolates showed negative results. Microscopic morphological identification of five selected isolates showed genus of each isolates, such as A.IV1.1 (Microclavia), D.III6.1 (Gliocladium), B.III6.1 (Periconia), A.II9.1 and D.II9.1 (Aspergillus).
