Abstract
Lactobacillus casei merupakan salah satu jenis probiotik yang memiliki gen tetM pengkode resistensi terhadap antibiotik tetrasiklin. Instabilitas gen tetM dapat diamati dari hasil adaptasi Lactobacillus casei yang dipapar pada konsentrasi subletal antibiotik tetrasiklin. Tujuan penelitian ini adalah untuk mengetahui tingkat resistensi Lactobacillus casei terhadap antibiotik tetrasiklin dan dampak pada instabilitas gen tetM. Hasil penelitian ini menunjukkan bahwa dosis letal L. casei adalah pada konsentrasi 60 µg.mL-1 dan dosis sub-letal dimulai dari konsnetrasi 10-50 µg.mL-1. Lactobacillus casei dengan kemampuan resistensi tertinggi didapatkan pada konsentrasi 70 µg.mL-1 yang ditandai dengan adanya perubahan fenotip yang merujuk pada terjadinya proses mutasi. Perubahan fenotip tersebut sudah dapat menjelaskan adanya instabilitas gen tetM akibat proses adaptasi. English Abstract
Lactobacillus casei has tetM gene which encodes antibiotic resistance to tetracycline. Gene instability of tetM gene can be observed through the result of adaptation processes in sub-lethal dose of tetracycline. The aim of this study was to determine the resistance level of Lactobacillus casei to the tetracycline exposure and its effect to the tetM Gene instability. The result of this study indicate that the lethal dose of lactobacillus casei is 60 µg.mL-1 and the sub-lethal dose is begin from 10-50 µg.mL-1. The highest concentration where Lactobacillus casei can survived is at concentration of 70 µg.mL-1, which characterized as the appearance of new phenotype changes. Changes in phenotype have been able to explain the instability of the tetM gene. It is proved that the exposure of sub-lethal dose can induce adaptation that lead to the mutation process. The most effective plasmid isolation method is Klaenhammer which resulting ~20 Kb of plasmid.
