Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit peradangan kronis pada saluran pencernaan, dengan penyebab pasti masih belum diketahui. IBD dapat disebabkan oleh induksi obat NSAID seperti indometasin secara berlebihan. Mekanisme kerja indometasin menghambat kinerja enzim siklooksigenase (COX), mengakibatkan kenaikan Reactive Oxygen Speciesi (ROS) serta aktivasi NF-κB pada jaringan lambung dan menimbulkan inflamasi. Air perak berperan sebagai antinflamasi, antimikroba, dan deteksi apoptosis. Air perak berpotensi dalam menekan inflamasi serta memperbaiki kerusakan organ lambung akibat penyakit IBD. Penelitian ini menggunakan tikus putih berjenis kelamin jantan,umur 8-12 minggu, dan berat badan 150-200 gram model IBD yang diinduksi dengan indometasin. Terdapat 5 kelompok perlakuan, yaitu kelompok negatif tanpa pemberian apapun, kelompok kontrol positif, P1, P2, dan P3 yang diinduksi indometasin 15 mg/kgBB diberikan satu kali sebelumnya, kemudian untuk kelompok P1, P2, dan P3 diterapi dengan air perak konsentrasi 25 ppm. Terapi air perak diberikan selama 10 hari secara sonde lambung. Parameter yang diamati dalam penelitian ini berupa akspresi NF-κB menggunakan metode imunohistokimia dan histopatologi lambung dengan pewarnaan HE (Hematoxyline-eosin). Analisa data hasil penelitian menggunakan metode ANOVA dan dilanjutkan uji tukey dengan tingkat kepercayaan α=0,05. Hasil penelitian menunjukkan bahwa pemberian terapi air perak dengan volume 1 mL, 2 mL, dan 3 mL pada tikus putih model IBD hasil induksi indometasin, dapat mengurangi ekspresi NF-κB, infilrasi sel radang, serta erosi epitel. Kesimpulan dari penelitian ini adalah terapi air perak dengan pemberian 3 mL dapat menurunkan ekspresi NF-κB, dan perbaikan gambaran histopatologi lambung berupa turunnya sel inflamasi, dan erosi epitel pada tikus IBD.
English Abstract
Inflammatory Bowel Disease (IBD) is a chronic inflammatory disease of the digestive tract, with the exact cause still unknown. IBD can be caused by excessive induction of NSAID drugs such as indomethacin. The mechanism of indomethacin action inhibits cyclooxygenase (COX), resulting in an increase in Reactive Oxygen Species (ROS) as well as the activity of NF-κB in gastric tissue and enhance proinflammatory cytokines production. Silver water acts as an anti-inflammatory, antimicrobial, and detection of apoptosis. Silver water has the potential to suppress proinflammatory cytokines and repair damage to gastric organs by IBD. This study used white male rats, age 8-12 weeks, and body weight 150-200 grams of the IBD model induced by indomethacin. There were 5 treatment groups, namely negative group without treatment, positive control, P1, P2, and P3 groups induced by indomethacin 15 mg / kgBW, on the next day P1, P2, and P3 groups were treated with 25 ppm of silver water. Via a gastric sonde for 10 days. The parameters observed in this study were NF-κB expression using immunohistochemistry and gastric histopathology methods with HE (Hematoxyline-eosin) staining. Analysis of the research data used the ANOVA method and continued with tukey test at α = 0.05. The results showed that administration of silver water therapy with a volume of 1 mL, 2 mL, and 3 mL in indomethacin-inducing white rats, can reduce the expression of NF-κB, infiltration of inflammatory cells and epithelial erosion. The conclusion of this study is silver water therapy by giving 3 mL can reduce the expression of NF-κB, and improve the histopathological picture of the stomach in the form of decreased inflammatory cells, and epithelial erosion in IBD white rats.
