Abstract
Tujuan dari penelitian ini adalah mengetahui pengaruf ekstrak kopi “Amstirdam” terhadap level Treg dan ekspresi Lp-PLA2 pada mencit diet tinggi lemak. Metode penelitian induksi HFD (High Fat Diet) pada semua mencti kecuali kelompok sehat selama 5 bulan dengan komposisi 8% protein dari kuning telur bebek, fruktosa 30%, 17% lemak sapi, kecuali 0,2% asam kolat yang hanya diberikan selama 2 bulan diawal induksi. Kemudian kelompok mencit treatment diberi esktrak kopi secara oral selama 2 minggu (DI = 104mg/kg, DII = 520mg/kg dan DIII = 5.200mg/kg). Mencit kemudian didislokasi dan organ spleen serta aorta diisolasi. Level Treg dan ekspresi Lp-PLA2 dianalisis dengan flowcytometry. Konfirmasi atherosclerosis dilakukan melalui pengamatan histologi aorta dengan metode paraffin dan pewarnaan H&E. Hasil analisis flowcytometri dilanjutkan dengan analisis statistik menggunakan SPSS 16.0 for windows dengan one-way ANOVA dengan nilai p= 0.05 dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa persentase ekspresi Lp-PLA2 mengalami penurunan setelah treatment dan nilai terendah pada kelompok treatment Dosis 2 yaitu 12,81% dibandingkan dengan kelompok Sehat dan Sakit yakni berturut-turut 9,75% dan 31,24% (p<0.05). Persentase Treg pada kelompok treatment Dosis 3 dan kelompok Sehat menunjukkan nilai yang tidak beda nyata pada uji Duncan yakni dengan nilai berturutturut 72,50% dan 69,43%, sedangkan nilai Treg pada kelompok sakit, Dosis 1 dan Dosis 2 berturut-turut adalah 33,82%, 58,33%, 50,57%. Pemberian ekstrak kopi amstirdam mampu menginduksi level Treg sehingga dapat mensupresi inflamasi pada aterosklerosi
English Abstract
The aim of this study is to determine the effect of "Amstirdam" coffee extract on Treg levels and Lp-PLA2 expression in mice induced High Fat Diet (HFD). The method of this research are HFD induction was performed on all mices except the healthy group for 5 months with diet composition, 8% protein from duck egg yolk, fructose 30%, 17% beef fat, except 0.2% colic acid which is only given for 2 months at the beginning of induction. Then the treatment mice group was given coffee extract orally for 2 weeks (DI = 104mg/kg, DII = 520mg/kg dan DIII = 5.200mg/kg). Mice were then dislocated and spleen and aorta organs isolated. Treg level and Lp- PLA2 expression were analyzed by flowcytometry. Confirmation of atherosclerosis is carried out through histological examination of the aorta with the paraffin method and H & E staining. The results of flowcytometry analysis continued with statistical analysis using SPSS 16.0 for windows with one-way ANOVA (p=0.05) and continued with Duncan test. The results showed that the lowest percentage of Lp-PLA2 expression (marker atherogenesis) in treatment group is Dosage 2 (12.81%) compared to the Healthy and Sickness group which was 9.75% and 31.24% respectively (p <0.05) . Based on the results of the Treg activation analysis, the values in the treatment group Dose 3 and the Healthy group showed values that were not significantly different from the Duncan test, with values of 72,50% dan 69,43% respectively. Whereas the Treg values in the sickness group, Dosage 1 and Dosage 2 were 33,82%, 58,33%, and 50,57% respectively. The increasing of Treg level caused by the treatment of coffee amstirdam extracts shows promising effect on atheroclerosis treatment.
