Abstract

Kanker merupakan penyakit seluler yang disebabkan oleh ketidaknormalan dari pembelahan sel. Sistem imun berperan dalam mengendalikan perkembangan sel-sel kanker. Keterlibatan beberapa sel T yang berperan dalam mekanisme tersebut yaitu sel T CD4+CD62L+, CD8+CD62L+ dan CD4+CD25+. Salah satu tanaman herbal yang dapat dimanfaatkan sebagai obat alternatif terhadap penyembuhan kanker yaitu ciplukan (Physalis minima L.). Tujuan penelitian ini mengetahui pengaruh pemberian ekstrak ciplukan (Physalis minima L.) terhadap jumlah relatif sel T CD4+CD62L+ CD8+CD62L+ dan CD4+CD25+ pada mencit (Mus musculus) betina model kanker payudara. Prosedur penelitian diawali dengan mempersiapkan mencit strain balb/c berumur 7-8 minggu. Perlakuan yang digunakan dosis ekstrak yang diberikan sebanyak 25 mg/kg BB, 50 mg/kg BB, 100 mg/kg BB, Kontrol Negatif, dan Normal. Mencit diinjeksi DMBA selama 6 minggu. Kemudian mencit diberi ekstrak ciplukan setiap hari selama 14 hari. Sel-sel imunokompeten diisolasi dari organ spleen kemudian dilakukan pewarnaan sel mengunakan antibodi dan dianalisis flow cytometry. Data di analisis dengan One-way ANOVA (Analysis of Variance) p=0,05 dilanjutkan dengan uji Post Hoc Tukey HSD menggunakan program SPSS versi 25.0 for windows. Hasil penelitian menunjukkan bahwa ekstrak ciplukan mampu meningkatkan jumlah relatif sel T CD4+CD62L+,CD8+CD62L+ dan menurunkan jumlah relatif sel T CD4+CD25+. Pemberian ekstrak ciplukan pada dosis 100 dan 50 mg/kg BB mampu meningkatkan Jumlah relatif sel T CD4+CD62L+ dan CD8+CD62L+ hingga mendekati normal. Pemberian dosis 100 mg/kg BB mampu menurunkan jumlah relatif sel T CD4+CD25+ hingga mendekati normal.

English Abstract

Cancer is a cellular disease that caused by abnormalities in cell division. The immune system plays an important role to control growth of breast cancer cells. The involvement of several T cells that play role in the mechanism are CD4+ CD62L+,CD8+ CD62L+ and CD4+CD25+ T cells. One of the herbs that can be used as an alternative medicine to cure cancer is ciplukan (Physalis minima L.). The purpose of this research is to know the effect of ciplukan extract on CD4+ CD62L+,CD8+ CD62L+ and CD4+CD25+ T cells in female mice (Mus musculus) breast cancer model. The research procedure begins with preparing 7-8 weeks old balb/c mice The treatment groups was divided into normal group, cancer group, and cancer group who were given ciplukan extract at a dose of 25 mg / kg, 50 mg / kg body weight and 100 mg / kg body weight. Mice were injected with DMBA for 6 weeks. Then, the mice were treated with ciplukan extract everyday for 14 days. Immunocompetent cells were isolated from the spleen organ and the cells will be stained using antibodies and analyzed using flow ctometry. Data are analyzed by using SPSS program, then tested with One-way ANOVA (Analysis of Variance) test p=0,05 Followed by the Post Hoc Tukey HSD test. The result show that ciplukan leaf extract can increased the relative amount of CD4+CD62L+ CD8+CD62L+ T cells and decreased the relative amount of CD4+CD25+ T cells. The treatment of ciplukan extract dose 100 mg/kg body weight and 50 mg/kg body weight was able to increased the relative amount of CD4+CD62L+ CD8+CD62L+ T cells and dose 100 mg/kg body weight was able to decreased the relative amount of CD4+CD25+ T cells to near normal treatment
