Abstract
Semanggi air (Marsilea crenata Presl.) memiliki kandungan fitokimia yang bersifat antioksidan di antaranya flavonoid, isoflavon, alkaloid, diterpenoid, vitamin C dan asam lemak. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol semanggi air terhadap ekspresi SOD1 dan Nrf2 pada organ testis tikus. Penelitian dilakukan pada empat kelompok tikus strain Wistar umur 3-4 bulan dengan berat badan antara 200-300 g. Kelompok kontrol (K) adalah kelompok yang tidak diberi perlakuan ekstrak etanol semanggi air dan ekstrak etanol semanggi air diberikan secara oral setiap hari selama 30 hari pada tiga kelompok perlakuan yaitu PS1, PS2 dan PS3 dengan dosis berturutturut 0,216 mg/gBB, 0,432 mg/gBB dan 0,648 mg/gBB. Pada hari ke- 31 tikus didislokasi dan diambil organ testisnya kemudian diisolasi sel dari organ testis untuk keperluan analisis persentase jumlah sel relatif yang mengekspresikan SOD1 dan Nrf2 menggunkaan flow cytometry. Data SOD1 diuji statistik Brown Forsythe dan uji lanjutan Games- Howell. Data Nrf2 diuji statistik one-way ANOVA dan diuji lanjutan Tukey HSD. Semua uji menggunakan signifikansi 0,05. Perlakuan pemberian ekstrak etanol semanggi air secara signifikan berpengaruh terhadap persentase jumlah sel relatif yang mengekspresikan SOD1 dan Nrf2. Pemberian perlakuan ekstrak etanol semanggi air dosis 2 (0,432 mg/gBB) secara signifikan meningkatkan persentase jumlah sel relatif yang mengekspresikan SOD1 dan dosis 1 (0,216 mg/gBB) meningkatkan persentase jumlah sel relatif yang mengekspresikan Nrf2 pada sel-sel penyusun organ testis dibandingkan dengan kelompok lainnya.
English Abstract
Water clover (Marselia crenata Presl.) contains phytochemicals with antioxidant property such as flavonoids, isoflavones, alkaloids, diterpenoids, vitamin C and fatty acid. This research aims to determine the effect of ethanol extract of water clover to the expression of SOD1 and Nrf2 in rat testis. The object of this research is four groups of 3-4 months old Wistar strain rats with 200-300 g of body weight. Control group (K) were not given the ethanol extract of water clover and ethanol extract of water clover were adminestered orally each day for 30 days to the treatment groups PS1, PS2 and PS3 with dosage as follow 0,216 mg/gBW, 0,432 mg/gBW and 0,648 mg/gBW. Rats were sacrificed at the day 31 by dislocation and the testis were dissected and cells were isolated for the analysis of percentage of relative cell that expressed SOD1 and Nrf2 using flow cytometry. Data of SOD1 were analyzed statistically by Brown Forsythe and followed by Games-Howell test. Data of Nrf2 were analyzed statistically by oneway ANOVA and followed by Tuckey HSD test. All statistic analysis were using α=0,05. The result showed that extract ethanol of water clover significantly affected the percentage of relative cell that expressed SOD1 and Nrf2. The administration of ethanol extract of water clover dosage 2 (0.432 mg/gBW) was significantly increased the expression of SOD1 and dosage 1 (0,216 mg/gBW) was increased the expression of Nrf2 in testicular cells.
