Abstract

Hair spray merupakan suatu produk kecantikan, dimana mengandung senyawa-senyawa berbahaya seperti akrilamida, formaldehida dan cocamide DEA. Apabila senyawa tersebut masuk kedalam organ limpa dan berinteraksi dengan sel dapat memicu terbentuknya radikal bebas. Penelitian ini dilakukan guna menganalisis pengaruh hair spray terhadap sifat kelistrikan, radikal bebas dan gambaran mikroskopis organ limpa pada mencit yang dipapari hair spray selama 30 hari. Sifat kelistrikan yang meliputi resistivitas, konduktivitas dan konstanta dielektrik diukur dengan mencari nilai kapasitansi dan resistansi menggunakan metode pasif dengan alat ukur multimeter. Impedansi diukur pada frekuensi 10 Hz hingga 100.000 Hz untuk masing-masing sampel. Electron Spin Resonance (ESR) dugunakan untuk mengidentifikasi radikal bebas dengan mengamati besar nilai frekuensi dan medan magnet. Pengamatan mikroskopis digunakan untuk melihat histologi organ limpa dengan pembuatan preparat. Pengukuran kadar Malondialdehyde (MDA) dilakukan sebagai parameter peroksidasi lemak yang diakibatkan oleh radikal bebas. Hasil dari penelitian menunjukan interaksi kandungan pada hair spray memicu terbentuknya radikal bebas anion superoksida (O₂⁻), semakin menurunnya nilai resistivitas, konstanta dielektrik, impedansi organ dan meningkatnya nilai konduktivitas dan kadar Malondialdehyde (MDA) pada organ seiring meningkatnya jumlah semprotan hair spray yang diberikan pada mencit.

English Abstract

Hair spray is a beauty product, which contains harmful compounds such as acrylamide, formaldehyde and cocamide DEA. When it enters spleen organ and interacts with cells, can caused the formation of free radicals. This research was conducted to analyze the effects of hair spray based on electrical properties, free radicals and microscopic images of mice's spleen organs that have exposed by hair spray for 30 days. The electrical properties which include resistivity, conductivity and dielectric constants were measured by looking for capacitance and resistance values using passive method with a multimeter as a measuring instrument. Impedance was measured by picoscope with variant frequency, from 10 Hz to 100,000 Hz for each sample. To identified the free radical, we used Electron Spin Resonance (ESR) by observing the value of the frequency and magnetic field of the samples. Microscopic observation was did to see the histology of the spleen organ by making prepared slides. Measurement of Malondialdehyde (MDA) levels was carried out as a parameter of fat peroxidation caused by free radicals which bounded to body fat. The results of this study showed that the interaction of content in hair spray caused the formation of superoxide anion free radicals (O₂⁻), decreasing values of resistivity, dielectric constant, organ impedance and increasing values of conductivity and Malondialdehyde (MDA) levels in the organ along with the increasing of hair spray's concentration.
