Abstract
Diabetes melitus (DM) merupakan penyakit metabolik yang berkaitan dengan peningkatan glukosa darah secara kronik dan kelainan kerja insulin. Bahan diabetogenik yang digunakan untuk menghasilkan hewan model diabetes tipe-1 dan tipe-2 dalam penelitian adalah streptozotocin (STZ). Sukrosa digunakan untuk memperparah kondisi hiperglikemi dan kerusakan sel β pankreas. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian diet tinggi sukrosa dan induksi Multiple Low Dose STZ terhadap kadar CD8 dan histopatologi ginjal pada tikus model diabetes melitus tipe 2. Hewan coba penelitian eksperimental ini berupa tikus putih (Rattus norvegicus) strain wistar jantan, berat badan 180-200g dibagi menjadi empat kelompok yaitu kelompok kontrol negatif (K-), Kelompok injeksi (K+) Multiple Low Dose STZ dosis 30 mg/kg BB, Kelompok injeksi (P1) Multiple Low Dose STZ dosis 30 mg/kg BB dan sukrosa 20% .Kelompok injeksi (P2) Multiple Low Dose STZ dosis 30 mg/kg BB 2 kali pada hari ke-6 dan ke-7 dan sukrosa 30% selama 7 hari. Injeksi STZ diberikan pada hari ke-6 dan 7, sedangkan sukrosa peroral selama 7 hari Pengukuran CD8 dilakukan dengan uji flowcytometry sedangkan histopatologi ginjal dianalisa menggunakan pewarnaan hematoksilin eosin. Hasil pada penelitian ini adalah induksi STZ 30 mg/kg BB dan diet tinggi sukrosa 20% dan 30% tidak menunjukan perbedaan yang signifikan terhadap kadar CD8 ginjal sedangkan hasil pengamatan histopatologi ginjal menunjukan perubahan morfologi berupa nekrosis dan deskuamasi epitel. Kesimpulan dari penelitian ini adalah Multiple Low Dose STZ 30 mg/kg BB dan diet tinggi sukrosa 20% dan 30% pada tikus putih model diabetes melitus tipe 2 dapat digunakan untuk membuat hewan model diabetes mellitus tipe 2 dengan tidak merusak fungsi ginjal.
English Abstract
Diabetes mellitus (DM) is a metabolic disease that is associated with an increase in blood glucose in chronic and insulin abnormalities. The natural diabetogenic material used to produce animal models of type 1 and type 2 diabetes in the study was streptozotocin (STZ). Sucrose is used to aggravate hyperglycemic conditions and pancreatic β cell damage. The purpose of this study was to determine the effect of giving a high sucrose diet and induction of Multiple Low Dose STZ on CD8 levels and renal histopathology in rats of type 2 diabetes mellitus. Experimental research animals were male wistar strains (Rattus norvegicus), weight 180 -200g was divided into four groups namely negative control group (K-), injection group (K +) Multiple Low Dose STZ dose 30 mg / kg BB, injection group (P1) Multiple Low Dose STZ dose 30 mg / kg BB and sucrose 20% Injection group (P2) Multiple Low Dose STZ dose 30 mg / kg BB 2 times on 6th and 7th day and 30% sucrose for 7 days. STZ injection was given on days 6 and 7, while oral sucrose for 7 days Measurement of CD8 levels was carried out by flowcytometry test while kidney histopathology was analyzed by using staining of hematoxylin eosin. The results of this study were STZ induction of 30 mg / kg body weight and a high sucrose diet of 20% and 30% did not show a significant difference in renal CD8 levels whereas renal histopathological observations showed morphological changes in the form of necrosis and desquamation of the epithelium. The conclusion of this study is that Multiple Low Dose STZ 30 mg / kg body weight and a high sucrose diet of 20% and 30% in type 2 diabetes mellitus can be used to make type 2 diabetes mellitus models without damaging kidney function.
