Abstract
Inflammatory Bowel Disease (IBD) adalah penyakit yang disebabkan oleh banyak faktor salah satunya penggunaan obat anti inflamasi non-steroid seperti indometasin. Indometasin bekerja dengan menghambat prostaglandin sehingga gastrointestinal menjadi asam dan menghasilkan Reactive Oxygen Species (ROS) tinggi. Air alkali memiliki Oxidation Reduction Potential (ORP) tinggi dan pH basa dapat digunakan untuk mengurangi sifat asam dan ROS. Penelitian ini bertujuan untuk mengetahui pengaruh terapi air alkali terhadap perubahan histopat jejunum serta aktivitas amilase. Penelitian ini menggunakan model rancangan acak lengkap (RAL). Model tikus yang dipakai dalam penelitian ini adalah tikus (Rattus norvegicus) jantan berumur 8-12 minggu dengan berat 150-200 gram. Tikus dibagi menjadi 4 perlakuan yaitu tikus kontrol negatif, tikus yang diinduksi indometasin, tikus terapi 1 (induksi indometasin dan terapi dengan volume 1 ml/ekor), dan tikus terapi 2 (induksi indometasin dan terapi dengan volume 2 ml/ekor). Induksi indometasin dengan dosis 15 mg/kg BB dan terapi air alkali di lakukan dua kali per hari dalam satu minggu. Pengukuran aktivitas amilase dengan metode NelsonSomogyi secara spektrofotometri, hasil diuji dengan ANOVA dan uji lanjutan Tukey dengan α = 0,05. Pengamatan histologis dilakukan dengan pewarnaan HematoxylinEosin. Terapi air alkali terutama pada volume 2 mL/ekor mempengaruhi secara signifikan aktivitas amilase sebesar 61% dan memperbaiki histopatologi jejunum berupa perbaikan mukosa vili dan bertambahnya sel goblet tikus yang diinduksi indometasin. Kesimpulan pada pemberian air alkali sebanyak 2 mL/ekor merupakan volume efektif dengan menurunkan aktivitas amilase dan memperbaiki histopatologi jejunum.
English Abstract
Inflammatory Bowel Disease (IBD) is a diseases that caused by a lot of factors, one of the causative factor is indomethacin, a non-steroidal of antiinflammatory drugs (NSAIDs). Indomethacin works by inhibiting prostaglandin that create a condition on gastrointesinal to be an acidic and produce more Reactive Oxygen Species (ROS). Alkaline-Ionic water have a high Oxidation Reduction Potential (ORP) and base pH that can reduce the acidic properties effect and ROS. This study aimed to determine the effect of alkaline water therapy on jejunal histopathic changes and amylase activity. This research used method by Fully Randomized Design. The rats (Rattus norvegicus) were 8-12 weeks old rats that have 150-200 grams body weight. The treatment was divided in to 4 groups (I): negative control rats, (II): positive control rats with indomethacin-induce, (III): IBD rats which was alkaline water therapy with a volume of 1 ml/rat, and (IV): IBD rats which was alkaline water therapy with a volume of 2 ml/rat. The IBD rats were induced with indomethacin in oral at dosage of 15mg/KgBW in a day and treated with alkaline-ionic water twice per day in a week. Amylase activity was measured with Nelson-Somogyi and used spectrophotometry, the result was statistically analyzed with ANOVA and Tukey’s HSD post hoc test (α = 0,05). Histopathology of jejunum was stained using Hematoxylin-Eosin and analyzed descriptively. Treatment using 2 mL of alkaline water significally affected amilase activity up to 61% and improved jejunal histopathology by regenerating villous mucosa and increasing its goblet cells. It was conculed that the effective volume to decrease the amylase activity and repairing jejunal histopathology was treatment of 2 mL of alkaline water per rat.
