Abstract

Obesitas merupakan penyebab utama munculnya penyakit degeneratif diantaranyadiabetes mellitus, hiperkolesterolemia, dislipidemia, gangguan metabolik dan gangguan sistem kardiovaskuler sperti stroke, hipertensi, dan aterosklerosis . Kejadian obesitas berhubungan dengan peningkatan kadar kolesterol total dan penurunan kadar High Density Lipoportein (HDL). Penelitian ini menggunakan desain hewan coba obesitas jenis Sprague Dawley (SD) dengan metode pemberian diet High Fat High Fructose (HFHF) modifikasi AIN-93M dengan kandungan lemak sebesar 51,64% dan fruktosa 30%. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian diet HFHF terhadap kadar kolesterol total dan kadar HDL terhadap parameter kadar kolesterol total dan kadar HDL. Desain penelitian ini merupakan studi eksperimental menggunakan rancangan randomized control group post-test.Jumlah sampel pada penelitian ini sebanyak 30 ekor tikus SD yang dibagi menjadi dua kelompok perlakuan yaitu diet HFHF dan diet normal AIN-93M termodifikasi yang mendapat diet selama 17 minggu. Hasil penelitian menyatakan bahwa terdapat perbedaan yang signifikan pada asupan pakan tikus antara kelompok diet HFHFdan diet normal AIN-93M termodifikasi dengan nilai p=0,000 (t-test, p<0,05). Sedangkan pada perubahan berat badan tikus menunjukkan tidak terdapat perbedaan yang signifikan antara kelompok diet HFHFdan diet normal AIN-93M termodifikasi dengan nilai p=0,103 (t-test, p>0,05). Pada penelitian ini diet HFHF dapat memberikan perbedaan yang signifikan pada hasil pengukuran kadar kolesterol total p=0,038, tetapi tidak memberikan perbedaan yang signifikan pada kadar HDL tikus penelitian dengan nilai p=0,075.

English Abstract

Obesity is a major cause of degenerative diseases including diabetes mellitus, hypercholesterolemia, dyslipidemia, metabolic disorders and cardiovascular system disorders such as stroke, hypertension and atherosclerosis. Obesityphenomenon has correlation with an increase in total cholesterol levels and a decrease in levels of High Density Lipoportein (HDL). This study used the obesity trial animal type Sprague Dawley (SD) design with a method of AIN-93M modified High Fat High Fructose (HFHF) diet with a fat content of 51.64% and 30% fructose. This study aims to determine the effect of giving HFHF diet on total cholesterol levels and HDL levels on parameters of total cholesterol levels and HDL levels. The design of this study was an experimental study using a randomized control group post-test design. The number of samples in this study were 30 SD rats which were divided into two treatment groups namely the HFHF diet and the modified AIN-93M normal diet which received a diet for 17 weeks. The results showed that there were significant differences in rat feed intake between the HFHF diet group and the modified AIN-93M normal diet with a value of p = 0,000 (t-test, p <0.05). However, there was no significant difference in the increase in body weight between the HFHF diet group and the modified AIN-93M normal diet with p = 0.103 (t-test, p> 0.05). In this study the HFHF diet can provide a significant difference in the measurement results of total cholesterol levels p = 0.038, but for HDL level it showed insignificant difference on each treatment with a value of p = 0.075.
