Abstract

Latar Belakang: Penyakit kardiovaskuler merupakan penyakit penyebab kematian nomor satu di dunia. Penyebab tersering penyakit kardiovaskuler adalah aterosklerosis. Aterosklerosis merupakan respon keradangan kronis terhadap deposit lipoprotein pada dinding arteri. Oxidized Low Density Lipoprotein (ox-LDL) adalah molekul utama yang berperan pada aterosklerosis. LOX-1 merupakan reseptor oxLDL yang terdapat di endotel dan berfungsi memperantarai ambilan oxLDL. Bila oxLDL berikatan dengan LOX-1 akan menyebabkan disfungsi endotel, sehingga terjadi ekspresi MCP-1 pada endotel dan sel otot polos pembuluh darah. Saat ini telah ada beberapa penelitian vaksin untuk mencegah aterosklerosis, baik melalui imunisasi pasif dan aktif memberikan hasil yang baik. Dengan demikian, strategi pencegahan aterosklerosis menjadi memungkinkan. Metode: Penelitian ini merupakan penelitian ekperimental menggunakan postest only control group design yang dilakukan pada 28 ekor tikus jantan Rattus norvegicus galur Wistar selama 56 hari. Tikus dibagi 7 kelompok, yaitu: kontrol negatif yang diberi diet normal, kontrol positif yang diberi diet aterogenik, kelompok perlakuan yang diberi diet aterogenik dan injeksi subkutan protein LOX-1 dengan dosis 1 ng+alum (P1), 10 ng+alum (P2), 100 ng+alum (P3), 1.000 ng+alum (P4), dan alum saja (P5). Pada akhir penelitian, jaringan aorta tikus diambil untuk dilakukan perhitungan ekspresi MCP-1 pada sel endotel dan sel otot polos aorta dengan metode immunohistokimia. Hasil: Analisa uji One-Way ANOVA menunjukkan bahwa pemberian protein LOX-1 belum dapat menurunkan secara bermakna ekspresi MCP-1 aorta Rattus norvegicus galur wistar yang diberi diet aterogenik (p>0,05). Kesimpulan: Hasil penelitian ini menunjukkan bahwa pemberian protein LOX-1 belum dapat menurunkan secara bermakna ekspresi MCP-1 pada aorta Rattus norvegicus galur wistar yang diberi diet aterogenik.

English Abstract

Background: Cardiovascular disease is the number one cause of death in the world. The most common cause of cardiovascular disease is atherosclerosis. Atherosclerosis is a chronic inflammatory response to lipoprotein deposit in artery walls. Oxidized Low Density Lipoprotein (ox-LDL) is a major molecule that plays a role in atherosclerosis. LOX-1 is an oxLDL receptor present in the endothelium and serves to mediate oxLDL uptake. When oxLDL binds to LOX-1, it will cause endothelial dysfunction, resulting in MCP-1 expression in the endothelial and smooth muscle cells of the blood vessels. Currently there has been some vaccine research to prevent atherosclerosis, both through passive and active immunization give good result. Therefore, the prevention strategy of atherosclerosis becomes possible. Methods: This study was a laboratory experimental study using post test only control group design and performed for 56 days towards 28 male of Wistar strain's Rattus norvegicus. Rats were divided into 7 study groups. The first group was negative control group fed with normal diet, the second group was positive control group fed with atherosclerotic diet, while the third and the following groups were test groups and those groups were fed with atherogenic diet and protein LOX-1 with dosage of 1 ng+alum (P1), 10 ng+alum (P2), 100 ng+alum (P3), 1,000 ng+alum (P4), and alum only diet (P5), respectively. At the end of the study, Rat's aorta were harvested and examined to evaluate the expression of MCP-1 in endothelial and smooth muscle cells of aorta by using immunohistochemistry method. Results: Analysis of the One-Way ANOVA test showed that administration of LOX-1 protein couldn’t significantly reduce the expression of MCP-1 in the aorta of Wistar strain's Rattus norvegicus with atherogenic diet (p>0.05). Conclusion: These results indicated that administration of LOX-1 protein couldn’t significantly reduce the expression of MCP-1 in the aorta of Wistar strain's Rattus norvegicus with atherogenic diet.
