Abstract
Aterosklerosis merupakan reaksi inflamasi kronis yang diawali dengan hiperkolesterolemia yang memicu proses disfungsi sel endotel, menyebabkan reaksi inflamasi dan proliferasi sel otot polos serta penumpukan sel busa yang menyebabkan peningkatan ketebalan dinding aorta. Statin sebagai lini pertama pengobatan memiliki berbagai efek samping pada pemakaian jangka panjang. Alga coklat (Sargassum sp.) sebagai sumber daya hayati perairan berdasarkan penelitian terdahulu memiliki kandungan senyawa bioaktif yaitu phlorotannin, fucoidan, dan polifenol yang bersifat anti-aterosklerotik dan anti-inflamasi serta efek proteksi dari radikal bebas. Penelitian ini bertujuan untuk mengetahui efektivitas pemberian ekstrak alga coklat (Sargassum sp.) dalam menghambat peningkatan ketebalan dinding aorta pada tikus wistar dengan diet aterogenik. Penelitian ini menggunakan metode randomized post test only controlled group design dengan sampel 30 ekor tikus jantan galur wistar. Sampel dibagi menjadi 6 kelompok, yaitu perlakuan 1 (Kelompok Negatif), 2 (Kelompok Positif), 3, 4, dan 5 yang diberi diet aterogenik dan ekstrak alga coklat 150 mg/kgBB/hari, 300 mg/kgBB/hari dan 600 mg/kgBB/hari, serta kelompok 6 yang diberi simvastatin dosis 4 mg/kgBB/hari mulai dari minggu ke-5 penelitian sebanyak 1 kali per hari. Penelitian dilakukan selama 12 minggu dengan parameter ketebalan dinding aorta. Hasil uji statistik Kruskall-Wallis didapatkan nilai p = 0,672 (p > 0,05) dimana tidak terdapat perbedaan signifikan antar kelompok perlakuan. Dari hasil penelitian ini, maka dapat disimpulkan bahwa pemberian ekstrak alga coklat (Sargassum sp.) tidak efektif dalam menghambat peningkatan ketebalan dinding aorta pada tikus wistar model hewan aterosklerosis dengan diet aterogenik, sehingga disarankan untuk menambah durasi intervensi pada penelitian serta dengan penggunaan metode diet tipe yang lain.
English Abstract
Atherosclerosis is a chronic inflammatory reaction that begins with hypercholesterolemia which triggers the process of endothelial cell dysfunction, causes inflammatory and proliferative reactions of smooth muscle cells and buildup of foam cells which causes an increase in aortic wall thickness. Statins as first-line treatment have various side effects on long-term use. Brown algae (Sargassum sp.) As aquatic biological resources based on previous research contain bioactive compounds namely phlorotannin, fucoidan, and polyphenols as an antiatherosclerotic, anti-inflammatory and protective effects of free radicals. This study aims to determine the effectivity of giving brown algae extract in inhibiting an increase in aortic wall thickness in wistar rats with an atherogenic diet. This study used a randomized post-test only controlled group design method with a sample of 30 wistar strain male rats. Samples were divided into 6 groups, namely treatment 1 (Negative Group), 2 (Positive Group), 3, 4, and 5 who were given an atherogenic diet and brown algae extract 150 mg/KgBB/day, 300 mg/KgBB/day and 600 mg/kgBB/day, and group 6 who were given simvastatin dose 4 mg/KgBB/day starting from the fifth week of the study once per day. The study was conducted for 12 weeks with parameters of the aortic wall thickness. The Kruskall-Wallis statistical test results obtained p = 0.672 (p > 0.05) where there were no significant differences between treatment groups. From the results of this study, it can be concluded that the administration of brown algae extract (Sargassum sp.) was not effective in inhibiting the increase of aortic wall thickness in atherosclerotic animal model wistar rats with an atherogenic diet, so it’s recommended to increase the duration of intervention in the study and by using other types of diet methods.
