Abstract
Penyakit kulit merupakan salah satu penyakit yang sering dijumpai di negara beriklim tropis, salah satunya impetigo. Penyebab penyakit ini karena infeksi bakteri dan sering kali oleh Streptococcus pyogenes. Terapi empiris mulai dipertimbangkan akibat meningkatnya prevalensi bakteri yang resisten terhadap antibiotik. Rumput teki (Cyperus rotundus) tumbuh sebagai gulma dan diketahui mengandung senyawa alkaloid, tanin, saponin, flavonoid, dan masih banyak lagi yang umumnya berfungsi sebagai anti bakteri, anti tumor, anti kanker, dan anti alergi. Telah berkembang metode ekstraksi baru yaitu Microwave Assisted Extraction. Penelitian ini bertujuan untuk mengetahui efek antibakteri dari microwave assisted extraction rimpang rumput teki (Cyperus rotundus) terhadap Streptococcus pyogenes secara in vitro. Penelitian menggunakan desain eksperimentaI laboratorium melalui uji kepekaan antimikroba metode difusi sumuran. Konsentrasi ekstrak rimpang rumput teki yang digunakan 10%, 20%, 40%, 60%, 80%, dan 100% dengan pengulangan empat kali. Berdasarkan hasiI peneIitian, zona inhibisi terbentuk pada seluruh konsentrasi uji 10%, 20%, 40%, 60%, 80%, dan 100%. Analisa statistik One-Way ANOVA Test menunjukkan perubahan konsentrasi ekstrak rimpang rumput teki memiliki efek yang signifikan terhadap zona inhibisi pertumbuhan bakteri Streptococcus pyogenes (p<0.05). Uji korelasi Pearson menunjukkan adanya hubungan yang kuat (0.861) sehingga dapat disimpulkan semakin tinggi konsentrasi ekstrak rimpang rumput teki, maka semakin besar diameter zona inhibisi yang terbentuk. Berdasarkan hasil penelitian ini, dapat disimpulkan bahwa ekstrak rimpang rumput teki (Cyperus rotundus) memiliki efek antibakteri terhadap bakteri Streptococcus pyogenes secara in vitro.
English Abstract
Skin disease is one of the most common diseases in tropical climate countries, and one of them is impetigo which occurs due to bacterial infection and commonly caused by Streptococcus pyogenes. Empirical theraphy nowadays is starting to be reconsidered due to the evolution of bacteria which allow them to become more resistant to antibiotics. Cyperus rotundus or nut grass growth as a weed and contains alkaloid compounds, tanin, saponin, flavonoid, and many more that commonly can be used as anti-bacterial, anti-tumor, anti-cancer, and anti-allergy. Nowadays there is a new extraction method called Microwave Assisted Extraction. This research was a laboratoy experiment design using welldiffusion method and performed in order to know the antibacterial effect of nut grass rhizome which is extracted using the microwave assisted extraction method against Streptococcus pygones in vitro. The concentration of nut grass rhizome extract were 10%, 20%, 40%, 60%, 80%, and 100% with four repetition. The result showed the inhibition zones ware formed in all concentration 10%, 20%, 40%, 60%, 80%, and 100%. Statistical analysis using One-Way ANOVA Test showed a significant result in the concentration changes toward inhibition zone of Streptococcus pyogenes growth (p<0.05). Pearson correlation test showed there was a strong relationship with the positive direction (0.861) which could means that the higher concentration of nut grass rhizome extract, the larger inhibition zone which can be formed. Based on this in vitro research, it can be concluded that nut grass rhizome (Cyperus rotundus) extract has an antibacterial effect against Streptococcus pyogenes in vitro.
