Abstract

Beberapa penelitian membuktikan konsumsi diet tinggi lemak (DTL) dapat mengakibatkan gangguan organ reproduksi, selain diet tinggi lemak beberapa penelitian telah membuktikan bahwa pemberian penyedap rasa Monosodium Glutamate (MSG) dapat mempengaruhi organ reproduksi yang dapat menyebabkan infertilitas. Namun, dosis MSG yang digunakan melebihi dosis rata rata konsumsi harian manusia. Maka dari itu, penelitian ini akan melihat pengaruh kombinasi diet tinggi lemak dan MSG berbagai dosis yang disesuaikan dengan konsumsi harian manusia terhadap sel epitel sekretorik dan otot polos tuba falopi. Penelitian ini menggunakan tikus putih Rattus novergicus betina dengan usia 6-8 minggu dan berat 140-200 gram. Penelitian dilakukan di Laboratorium Fisiologi dan Patologi Anatomi Fakultas Kedokteran Universitas Brawijaya. Penelitian dilakukan selama 56 hari yang terdiri dari 6 kelompok yaitu kontrol negatif, DTL (K+1), MSG 0,7mg/gBB (K+2), DTL+MSG 0,05mg/gBB (PI), DTL+MSG 0,2 mg/gBB (PII), DTL+MSG 0,35mg/gBB (PIII). Pemberian MSG dilakukan peroral dengan sonde selama 56 hari dan pemberian diet tinggi lemak diberikan secara ad-libitum.Tikus dibedah pada hari ke 57- 61 pada saat tikus mencapai fase proestrus. Pada tikus yang belum mencapai fase proestrus pada lebih dari hari ke-61 akan dieksklusi. Pengamatan sel epitel sekretorik tuba falopi dilakukan oleh peneliti dengan 12 lapang pandang dan pengukuran ketebalan otot polos tuba falopi dilakukan oleh peneliti dengan 12 titik. Hasil uji one way ANOVA, rerata jumlah sel epitel sekretorik (p=0,008) dan ketebalan otot polos tuba falopi (p=0,001) pada kelompok yang diberikan kombinasi diet tinggi lemak dan MSG dosis rendah menurun secara signifikan jika dibandingkan dengan kontrol negatif. Namun tidak signifikan jika dibandingkan dengan kontrol positif diet tinggi lemak saja (K+1) dan MSG dosis tinggi 0,7mg/Gbb (K+2). Hasil penelitian ini membuktikan bahwa pemberian MSG dosis rendah jika dikombinasikan dengan diet tinggi lemak akan menghasilkan penurunan rerata jumlah sel epitel sekretorik dan penipisan ketebalan otot polos tuba falopi yang sama dengan pemberian MSG dosis tinggi. Pemberian kombinasi diet tinggi lemak dan MSG dapat mengakibatkan gangguan hormonal pada tikus yang mengakibatkan terganggunya fungsi tuba falopi.

English Abstract

Several studies have proved that the consumption of a high-fat diets (HFD) can lead to disorders of the reproductive organs. In addition to high-fat diets, some research also proved that the given of flavor enhancer, Monosodium Glutamate (MSG), can affect reproductive organs to raise the possibility of infertility. However, the mentioned cases occur when the doses of MSG used exceeds the average daily consumption of a human. Therefore, this research will be focused at the effect of the combination of various doses in High Fat Diets and MSG, which adjusted to human’s needs in daily basis, against the secretory epithelial cells and smooth muscle of fallopian tubes. The object used in this research was a white rat Rattus novergicus females in age of 6-8 weeks and weighed of 140-200 grams. Research was done in the Laboratory of Physiology and Pathology of Anatomy, Faculty of Medicine University of Brawijaya. The research was conducted in 56 day long consisting of 6 groups: the negative control, HFD (K + 1), MSG 0, 7mg/gBB (K + 2), HFD + MSG 0, 05mg/gBB (PI), HFD + MSG 0.2 mg/gBB (PII), HFD + MSG 0, 35mg/gBB (PIII). The MSG supply is done with sonde via oral for 56 days and high-fat diets supply is given in ad-libitum. Rat is dissected on day 57 - 61 once they reached the proestrus phase. The rats which had not be in the proestrus phase on day 61 will be excluded. Observations of secretory epithelial cells of fallopian tubes are done by researchers with 12 views and smooth muscle thickness measurement of fallopian tubes is done by 12 points. One way ANOVA test result shows the average amount of secretory epithelial cells (p = 0.008) and smooth muscle thickness of fallopian tubes (p = 0.001) in the group that was given a low dose combination of a high-fat diets and MSG is significantly decreased compared to the negative control. Yet, it was not significant enough when compared to the high-fat diet of positive controls (K + 1) and MSG high doses 0, 7mg/Gbb (K + 2). In the conclusion, the study proved that low dose of MSG supply when combined with a high-fat diet will result in the same decreasing of the average number of secretory epithelial cells and the thinning of smooth muscle thickness of the fallopian tubes with administering the high doses of MSG. The effect of a combination a high-fat diet and MSG can cause hormonal disturbances in rats that resulted in disruption of the functions of the fallopian tube.
