Abstract
Enzim selulase dimanfaatkan sebagai biokatalisator untuk menghasilkan produk yang lebih bernilai seperti fermentasi biomassa dan proses hidrolisis. Namun, enzim ini masih sulit didapatkan di Indonesia dan juga memilki harga yang relatif tinggi. Enzim selulase dapat diproduksi dari mikrofungi selulotik seperti salah satunya fungi Trichoderma harzianum. Produksi enzim selulase dilakukan melalui proses fermentasi padat dan didukung oleh adanya substrat sebagai media sekresi enzim. Substrat pada proses produksi enzim pada penelitian menggunakan substrat dari buah bintaro (Cerbera odollam) yang diketahui mengandung tinggi lignoselulosa sehingga memiliki potensi untuk proses sintesis enzim selulase. Kandungan lignoselulosa pada buah bintaro memilki rincian yaitu selulosa sebesar 36,945% dan lignin sebesar 38% (Handoko, 2012). Rancangan penelitian yang digunakan adalah Rancangan Acak Kelompok Faktorial yang disusun secara faktorial dengan dua faktor, faktor I merupakan lama waktu inkubasi yaitu 144 jam, 168, jam, dan 192 jam. Faktor II merupakan konsentrasi inokulum pada massa substrat yaitu 20%, 14% dan 11%. Masing-masing kombinasi perlakuan kemudian diulang sebanyak tiga kali, sehingga menghasilkan total 27 sampel. Data kualitas enzim yang dihasilkan kemudian akan dilakukan uji dengan Uji Kualitatif dan Uji Kuantitatif enzim. Selain itu pada proses pretreatment juga akan dilakukan uji kadar air bahan substrat agar prosedur yang dilakukan sesuai dan menimalisir tingkat kontaminasi.
English Abstract
Cellulase enzymes are used as biocatalysts to produce more valuable products such as biomass fermentation and hydrolysis processes. However, this enzyme is still difficult to obtain in Indonesia and also has a relatively high price. Cellulase enzymes can be produced from cellulotic microfungi such as Trichoderma harzianum. Cellulase enzyme production is done through solid fermentation process and supported by substrates as media of enzyme secretion. The substrate in the enzyme production process in the study used a substrate of Sea Mango fruit (Cerbera odollam) which is known to contain high lignocelluloses and thus has the potential for cellulase enzyme synthesis. The content of lignocellulose in bintaro fruit has details of cellulose of 36.945% and lignin of 38% (Handoko, 2012). The research design used was Randomized Block Design which was arranged factorially with two factors, the first factor was incubation time of 144 hours, 168 hours and 192 hours. The second factor is the concentration of inoculum of mass substrate that is 20%, 14% and 11%. Each treatment combination was repeated three times, resulting in a total of 27 samples. The resultant quality of enzyme will then be tested with Qualitative and Quantitative Test of enzyme. In addition to the pretreatment process will also be tested for moisture content of substrate materials for the appropriate procedures and minimize the level of contamination.
