Abstract
Perairan laut Indonesia kaya akan potensi mikrobia laut. Saat ini mikrobia potensial di lautan kita yang belum banyak dimanfaatkan adalah khamir laut. Khamir laut potensial untuk dimanfaatkan diberbagai bidang, salah satunya industri makanan, fermentasi, riset biomedik, farmakologi, teknologi lingkungan dan pengembangan sains. Tetapi hingga kini di Indonesia belum ada bank data hasil penelitian khamir laut. Penelitian ini bertujuan untuk mendapatkan data molekular tentang profil pita DNA ribosom isolat khamir laut yang teramplifikasi pada daerah ITS dan untuk mengetahui identitas isolat khamir laut berdasarkan profil pita DNA ribosom yang teramplifikasi pada daerah ITS. Penelitian telah dilaksanakan di Laboratorium mikrobiologi Fakultas Perikanan dan Laboratorium Rekayasa Genetika, Balai Pengkajian Bioteknologi –BPPT, Serpong, Tangerang pada bulan Juni sampai Oktober 2009. Tahapan penelitian ini meliputi : pemurniaan biakan khamir laut, pengamatan morfologi, isolasi DNA total dan amplifikasi DNA ribosom pada daerah ITS ( Internal Transcribed Spacer ). Hasil penelitian didapatkan satu isolat khamir laut yang memiliki ukuran panjang 9-28 µm dan lebar 1,5-3 µm memiliki hifa tidak bersekat, berwarna krem transparan, terdapat miselium sejati dan tidak ada pseudomycelium . Pertumbuhan vegetatifnya berupa pertunasan multilateral. Selanjutnya h asil amplifikasi DNA dengan teknik PCR menggunakan primer ITS1 dan ITS4 terhadap isolat khamir laut menghasilkan satu pita dengan ukuran berkisar antara 500-750 bp.
English Abstract
Indonesian seas are rich of sea microbes potensial. One of re that have not been more used yet is marine yeast. Marine yaest is potensial to be used in some field such as food industrie, fermentation, biomedical research, pharmacology, environmental technology and science development. Unfortunately, Indonesia does not have yet any data bank of research result on marine yeast. objective of this research are to know profil fragmen ribosomal DNA of marine yeast that amplification of ITS region and to know identity isolate of marine yeast base on profil fragmen ribosomal DNA amplification of ITS region. This research has been conducted in Microbiology Laboratory, Marine Biotechnology Laboratory of Fisheries Faculty at Brawijaya University, and Technology Genetic Laboratory, Agency for Assessment and Aplication of Technology-BPPT, Serpong, Tangerang on Juni 2009 up to October 2009. research stages involved marine yeast purifiying, morphology observation, isolation of total DNA and yeast ribosomal DNA amplification of ITS ( Internal Transcribed Spacer ) region. result of this research show that one isolate of marine yeast , that have size is 9-28 µm long and 1,5-3 µm wide. It has unseptated hypha, transparent cream colour, has true mycelium and not have pseudomycelium. Its vegetative reproduction is multilateral budding. result of amplification ribosomal DNA with PCR used primer ITS1 and ITS4 to marine yeast isolate to produced single band DNA with 500-750 bp range.
