Abstract
Tujuan dari penelitian ini adalah untuk mengevaluasi pakan terkontaminasi dioksin pada kinerja produksi, telur, hati, konsentrasi dioksin lemak perut, dan ekspresi gen di hati ayam Red-Fea Red Country Chickens. Jumlah total 32 ayam peletakan dibagi menjadi 2 kontrol perawatan dan kelompok dioxin. Enam belas ayam diberi makan dengan pakan dioksin kontaminasi selama empat belas hari, dilanjutkan dengan pakan bersih hingga 42 hari. Ayam dikorbankan pada 2, 14, 28, dan 42 setelah perawatan dioksin. Dosis 17 Dixin Congener ditambahkan ke diet adalah 5,5 ng TEQ KG-1 Feed. Data eksperimental dianalisis dengan desain faktorial 2 x 4. Perbedaan antara perawatan dianalisis dan dibandingkan dengan rata-rata kuadrat dan nilai p 0,05 dianggap signifikan. Hasil menunjukkan bahwa Dioksin secara signifikan mengurangi rata-rata harian (ADG) 5,76 g (p <0,05). Dioksin diadministrasikan tidak berbeda secara signifikan (p 0,05) dalam berat hati, persentase hati, berat limpa, persentase limpa, berat telur dan produksi telur. Namun, perawatan dioksin cenderung mengurangi berat limpa relatif (p = 0,08). Konsentrasi PCDD / PCDF dalam hati lebih tinggi dari kelompok kontrol. n, konsentrasi PCDD / PCDF di hati secara bertahap menurun setelah pakan terkontaminasi dioksin diubah dengan umpan bersih. Hati adalah konsentrasi dioxin tertinggi di jaringan, dilanjutkan dengan telur, dan lemak perut. Dioksin mengubah ekspresi gen metabolisme lipid di hati. Selain itu, dioksin mengubah ekspresi gen di hati yang terkait dengan jalur pensinyalan ppar, jalur metabolisme piruvat dan jalur glikolisis / glukoneogenesis. Gen yang memiliki peran dalam jalur pensinyalan ppar adalah apolipoprotein AV (APOA5), carnitine palmitoyltransferase 1a (hati) (cpt1a), cytochrome p450 keluarga 7 subfamily a, polipeptide 1 (CYP7A1), matriks metallopeptidase 1 (mmp1) (MMP1), dan fosfoenolpyruvate carboxykinase 1 (PCK1). Gen yang memiliki peran dalam metabolisme piruvat dan jalur glikolisis / glukoneogenesis adalah ACYL-COA SYN SES anggota keluarga rantai pendek 1 (acss1), fosfoenolpyruvate carboxykinase 1 (pck1), piruvat dehydrogenase (lipoamide) alpha 1 (PDHA1), dan mirip dengan asetil -COA SYN TASE 2-LIKE (LOC423347).
English Abstract
aim of this research was to evaluate dioxin contaminated feed on production performance, egg, liver, abdominal fat dioxin concentration, and gene expression in liver of red-fea red Taiwan country chickens. A total number of 32 laying hens were divided into 2 treatments control and dioxin group. Sixteen chickens were fed with contamination dioxin feed for fourteen days, were continued by clean feed up to 42 days. Chickens were sacrificed at 2, 14, 28, and 42 after dioxin treatment. dose of 17 dioxin congeners were added to diet is 5.5 ng TEQ kg-1 feed. Experimental data was analyzed with 2 x 4 Factorial Design. Differences between treatments were analyzed and compared by least squares mean and a P-value of 0.05 was considered significantly. result indicated that dioxin was significantly reduced average daily gain (ADG) 5.76 g (P < 0.05). Dioxin administrated was no significantly different (P 0.05) in liver weight, liver percentage, spleen weight, spleen percentage, egg weight and egg production. However, dioxin treatment tended to decrease relative spleen weight (P = 0.08). PCDD/PCDFs concentrations in liver were higher than control group. n, concentration PCDD/PCDFs in liver gradually decreased after dioxin contaminated feed was changed by clean feed. Liver is highest concentration of dioxin in tissue, continued by egg, and abdominal fat. Dioxin altered lipid metabolism gene expression in liver. Moreover, dioxin altered gene expression in liver which related with PPAR signaling pathway, pyruvate metabolism pathway and glycolysis/gluconeogenesis pathways. Genes that has role in PPAR signaling pathway are apolipoprotein A-V (APOA5), carnitine palmitoyltransferase 1A (liver) (CPT1A), cytochrome P450 family 7 subfamily A, polypeptide 1 (CYP7A1), matrix metallopeptidase 1 (interstitial collagenase) (MMP1), and phosphoenolpyruvate carboxykinase 1 (PCK1). Genes that has role in pyruvate metabolism and glycolysis/gluconeogenesis pathways are acyl-CoA syn ses short-chain family member 1 (ACSS1), phosphoenolpyruvate carboxykinase 1 (PCK1), pyruvate dehydrogenase (lipoamide) alpha 1 (PDHA1), and similar to acetyl-CoA syn tase 2-like (LOC423347).
