Abstract

Uji elektrokimia yang sangat sensitif untuk deteksi DNA spesifik urutan telah dikembangkan dengan mengintegrasikan 3 teknik. Menggunakan label barcode DNA pada lateks microsfer (lateks. diameter sekitar 468 nm), amplifikasi perak dan asam nukleat peptida (PNA) sebagai probe tangkap. Label lateks barcode DNA diukur, terdiri dari reporter probe DNA dan banyak DNA barcode yang diumumkan, menyediakan jumlah besar barcode DNA pada partikel lateks. Amplifikasi perak dikembangkan, dengan peluang besar untuk meningkatkan sinyal elektrokimia, dan dilakukan oleh cluster logam perak secara elektrostatik terikat dengan lateks barcode DNA. PNA oligomers menyediakan selektivitas afinitas-afinitas tinggi dan menghilangkan sinyal latar belakang dari cluster perak non spesifik pada permukaan elektroda. Oligomer PNA yang diimobilisasi pada Layar Dicetak Elektroda (SPES) mengikat target komplementer DNA dan Label Lateks DNA barcode, kemudian mengembangkan peningkatan perak, diikuti oleh pembubaran asam dan ion perak diukur dengan voltametri pengupasan anodik (ASV). Pengaruh variabel eksperimental yang relevan, termasuk konsentrasi PNA, durasi hibridisasi, suhu dan peningkatan peningkatan, diperiksa dan dioptimalkan. Dengan menggunakan PNA sebagai probe penangkapan dapat mengurangi latar belakang. Amplifikasi sinyal dengan menurunkan batas deteksi serendah 210 zm dicapai dan menunjukkan selektivitas yang mengesankan dengan diskriminasi yang berhasil dari DNA target dari urutan ketidakcocokan tunggal dan non-komplementer. Arsitektur biosensor DNA elektrokimia novel ini dapat terbukti cocok untuk deteksi target dan bakteri DNA genom tanpa pretreatment tetapi tidak memberikan sensitivitas yang lebih baik, itu dikaitkan dengan komponen kompleks genomik yang dapat menghambat proses hibridisasi.

English Abstract

A highly sensitive electrochemical assay for the sequence-specific DNA detection has been developed by integrating 3 techniques; using DNA-barcode labeling on latex microsphere (latex; diameter about 468 nm), silver amplification and peptide nucleic acids (PNA) as capture probe. DNA barcode latex labeling measured, composed of reporter probe DNA and many aminated barcode DNA, provides the huge amount of DNA barcode on latex particle. Silver amplification was developed, with a great opportunity to increase electrochemical signal, and performed by silver metal cluster electrostatically bound to the DNA barcode latex. PNA oligomers provide the high affinity-sequence selectivity and eliminate background signal from non specific silver cluster on the electrode surface. The immobilized PNA oligomers on screen printed electrodes (SPEs) bind to complementary target DNA and DNA barcode latex label, then developed silver enhancement, followed by acid dissolution and measured silver ions by anodic stripping voltammetry (ASV). The influence of relevant experimental variables, including the PNA concentration, the duration of the hybridization, temperature and enhancement step, is examined and optimized. By using PNA as a capture probe can reduce the background. Signal amplification with lowering of the detection limit as low as 210 zM were achieved and exhibited impressive selectivity with successful discrimination of the target DNA from single mismatch and non complementary sequence. This novel electrochemical DNA biosensor architecture may prove suitable for the detection of target and genomic DNA bacteria without pretreatment but does not give better sensitivity , it attributed to complexes component of genomic that could inhibit hybridization process .
